摘要
将成熟的禽流感间接ELISA快速诊断技术试剂盒化 ,确定其外包装、规格、盒内组成 ,对盒内各组分的性状、保存、质量控制以及诊断试剂盒的特异性、敏感性、可重复性、符合率、与国外同类产品比较、保存期等进行了全面、详细的研究。试剂盒由 9种成套试剂组成 ,在 - 15℃至 - 2 0℃保存 5 40天各项性能仍然很好。该试剂盒与进口禽流感间接ELISA诊断试剂盒对同样血清样品检测 ,符合率为 10 0 %。拥有该试剂盒 ,只需准备生理盐水 ,即可对大量血清样品同时进行检测 ,操作简单方便、结果特异性强、敏感性高、稳定、快速 ,更适合于禽流感免疫抗体的监测及现地疫病诊断等需要。先后制备诊断试剂盒 16个批次 ,已成功应用于我国省、市兽医站、兽医研究所、农业院校、农科院、农业公司、口岸检疫部门及鸡场等各领域对禽流感的定性诊断、流行病学调查、免疫抗体监测等。
An enzyme_linked immunosorbent assay (ELISA) test kit employing recombinant nucleoprotein expressed in baculovirus system was developed for detection of antibodies against avian influenza virus(AIV).The kit was composed of eight reagents and the reagents were stablewell when storing at 15℃～-20℃ for 540 days. The variant coefficients of the OD value within assay and between assay ranged from 4.3 to 7.2, and from 4.6 to 9.8% respectively. 5 field_origin sera and 7 positive reference sera were evaluated by the kit of ours and that of U.S.A. The correspondence between ELISA kit of ourselves and from U.S.A was 100% when the same sera sampleswas 1:400.The kit hasbeen successfully used in at least 11 provinces and cities in chinaup to now.Only normal saline was necessary with the kit and a large numberof sera samplescould be tested at a time and the positive result could be reported within 1.5 hours. The results demonstrated that thetest kit was fit for all fields to detect and diagnose AIV antibodies in chicken sera.
