摘要
目的 为了探讨血清高密度脂蛋白胆固醇 (HDL C)各种测定方法的优劣性 ,选择更加符合临床要求的方法。方法 磷钨酸镁沉淀法 (PTA Mg2 + 法 ) ,改良磷钨酸镁沉淀法 (简称改良法 ) ,聚乙二醇沉淀法 (PEG法 ) ,选择性遮蔽法 (直接测定法 )。结果 各种方法的批内精密度除PEG法外均小于 5 % ,其中直接法小于 2 % ;批间精密度除直接法小于 2 %外 ,余均大于 5 %。直接法、改良法与PTA Mg2 + 法测定结果一致性好 ,其相关系数r分别为 0 988和 0 990。在HDL C浓度为 2 85mmol/L范围内各种方法的线性均良好。结论 PTA Mg2 + 法准确性基本符合临床要求 ,成本低 ,适合手工分析。直接法简便快捷 ,易操作 。
Objective To discuss the advantage and shortage of different methods for measuring high density lipoprotein cholesterol, choose the best one for clinical application.Methods Phosphotungstic acid Magnesium chloride(PTA Mg 2+ )precipitation method, Amended phosphotungstic acid Magnesium chloride precipitation method(amended method),Polyeglycol(PEG) precipitation method, Homogeneous assay(direct measurement).Results Varieties of methods within day variance were<5%, except for PEG method,only direct measurement method′s within day variance was <2%.The day and day variance were>5%,except for direct measurement(<2%).Amended method and direct measurement were in good agreement with PTA Mg 2+ (r=0 990 and 0 988). All methods were linear up to at least 2.85mmol/L.Conclusion PTA Mg 2+ method is coincident with clinical requirement and is low cost, and it is suitable for routine manual analysis.Direct measurement method is simple, rapid, easy to handle , and allows full automation.
