大麦条纹花叶病毒新疆株RNA5′-末端帽子结构的鉴定 被引量:2

IDENTIFICATION OF RNA 5' CAP STRUCTURE FROM BARLEY STRIPE MOSAIC VIRUS ( XINJIANG STRAIN )

摘要 在弱碱条件下一些RNA病毒蛋白外壳可以特异性地从RNA5'-末端开始剥落,对大麦条纹花叶病毒新疆株(BSMV-XJ)RNA以含帽子结构的TMV-RNA和不含帽子结构的CpMV-RNA为正,负对照,在弱碱条件下剥壳,用合成的pCp特异性地标记RNA5'-末端帽子结构上的3'-羟基,经碱水解后纸电泳分离单核苷酸,见BSMV-XJ-RNA和TMV-RNA同样含有m^7G~*p,而CpMV-RNA则无,证实BSMV-XJ-RNA在5'-末端具有m7G帽子结构,同时对BSMV的蛋白外壳在弱碱条件下剥落的时间,条件及标记方法进行了研究。 The coat proteins of some rod-shaped, RNA-containing plant viruses can be specifically stripped off from their RNA 5'-terminals under alkali condition. The 5'-terminal structure of BSMV-XJ-RNAs was compared with the m7G structure of TMV-RNA as a positive control and with the Vpg structure of CpMV-RNA as a negative one.After stripping off the coat proteins of the viuses in alkali solution the 3'-OH of the cap ware labelled by the synthetic pCp and then the incompletely labelled viruses were precipitated by PEG. The exposed nucleic acid of the partially stripped virions were hydrolyzed by alkali into single nucleotides which were monitered by UV light and autoradiography after paper electrophoresis. It is concluded that BMSV-XJ-RNA has the same m7G cap structure as the TMV-RNA's.
