Study on Rectal Mucosa Absorption Effects of Cobra Neurotoxins by ^(125)I Tracer Labelling Methods

摘要 目的 :建立测定家兔血浆中眼睛蛇神经毒素 ( Neurotoxins,Nt)的放射性核素 ( 1 2 5I)示踪法 ,观察冰茶栓中 Nt透直肠粘膜吸收情况。方法 :1 2 5I-Nt采用氯胺 T法标记 ,血浆中 Nt浓度采用同位素结合三氯醋酸 ( TCA)沉淀法测定。结果 :Nt浓度在5~ 5 0 0 ng/ml之间线性关系良好 ,相关系数 r=0 .9990 ,平均回收率 =86.8% ,RSD小于 1 0 % ,最小检测线为 3ng/ml。冰茶栓中 ,家兔按 0 .39mg/kg直肠给 1 2 5I-Nt后 ,能检测到 Nt在血浆中含量。结论 :放射性核素 ( 1 2 5I)示踪法可用于血浆中 Nt含量测定 ,冰茶栓中 Nt能透过直肠粘膜吸收。 To establish radioisotope( 125 I)tracer labelling methods for the determination of rectal mucosa absorption effects of Cobra Neurotoxins.Methods 125 I was labelled with chloranine-T method.Concentration of Nt in rabbit plasma was determined by radioisotope tracer labelling method in conjunction with trichloroacetic(TCA).Results Assay linearities were determined in the range of 5~500ng/ml in plasma(r=0 990),the average recovery was 86 8%.RSD was less than 10%,the lower limits of detection for Nt in plasma were 3ng/ml.After treated with 0 39mg/kg 125 I Nt in Bing Cha Shuan by rectal delivery,the concentration of Nt can be determined in plasma.Conclusion Radioisotope(125I)tracer labelling method is reliable and suitable for the determination of Nt in plasma,Nt in Bing Cha Shuan can be absorbed by rectal delivery.
