摘要 目的 :探讨乙醇对胃乙醇脱氢酶 (Alcohol dehydrogenase,ADH)活性变化的影响。方法 :Wistar大鼠 5 2只随机分成两组 :模型组 32只 ,对照组 2 0只。用乙醇直接灌胃的方法建立大鼠模型 ;用酶组织细胞化学染色技术 ,观察胃ADH活性变化 ,并用 L UZEX- F灰度图像分析仪进行半定量。结果 :随着乙醇灌注量的增加和灌注时间的延长 ,胃 ADH活性进行性下降 ,与对照组有显著性差异 (P<0 .0 5 ) ;停止灌注后 1个月 ,胃 ADH活性明显恢复 ,且与正常对照组无显著性差异 (P>0 .0 5 )。结论 :乙醇导致胃 ADH活性进行性下降 ,从而降低了乙醇首过代谢 。 Purpose: Effect of alcohol on activity variety of gastric alcohol dehydrogenase(ADH) was investigated. Methods: Fifty two rats were divided into two groups in model one(32/52) and control one (20/52), at random. Model one was made of directly perfusing alcohol into stomach. The activity varieties of gastric ADH were observed by the optical microscopy after gastric tissues were stained by enzyme histo cytochemistry assay. The activity varieties were determined by LUZEX F analysis instrument as a semi quantitative method. Results: It was shown that the activities of gastric ADH gradually decreased along with the volume of alcohol increased and the duration of perfusion prolonged. There were significant differences between model group and control one(P<0.05). The activity of gastric ADH significantly recovered one month after abstinence from alcohol. There were not significant differences between two groups(P> 0.05 ). Conclusion: Alcohol resulted in activity descending of gastric ADH, as a result of decreasing first pass metabolism of gastric alcohol and increasing alcoholic poisonous roles in liver and other organs.
