摘要
目的从内皮素ET、降钙素基因相关肽CGRP、多巴胺DA、去甲肾上腺素NE及肾上腺素E的变化方面研究益元活血丹、大黄及血栓心脉宁对老龄大鼠脑缺血再灌注胃肠损伤防护作用的机制。方法老龄20月龄以上大鼠分为正常对照组、模型组、尼莫地平组、益元活血丹大剂量及小剂量组、大黄组、血栓心脉宁组,观察大鼠全脑缺血30min再灌注60min后ET、CGRP、DA、NE、E含量的变化。结果老龄模型组ET高于老龄对照组,益元活血丹大剂量组和血栓心脉宁组及大黄组ET的降低和益元活血丹小剂量组CGRP水平升高均较老龄模型组差异显著。老龄模型组大鼠交感-肾上腺系统兴奋增强用药各组交感-肾上腺系统兴奋减低,以大黄组和益元活血丹组为明显。结论交感-肾上腺系统兴奋增强和CGRP-ET平衡失调参与老龄大鼠脑缺血再灌注胃肠损伤。各组药物对脑缺血再灌注胃肠损伤的保护作用机制可能涉及其抑制交感-肾上腺系统兴奋增强和调节CGRP与ET间的平衡失调。
To study thy protective effects of Yiyuanhuoxuedan(YHD),rhubarb and Xueshuanxinmaining(XXN) on gastrointestinal tract injury after brain ischemia reperfusion in the aged rats from the change of endothelin(ET),Dopamine(DA),Noradrenalin(NE)and Epinephrine(E),calcitonin gene related peptied (CGRP).Methods:Aged rats (older than 20 months) were divided into aged model group(AMG) and aged control group(ACG),nimodipine group(NG),high doses group and low doses group of YHD(HYHD,LYHD),rhubarb group(RG),XXN group(XXNG).It was measured that the contents of ET,CGRP,DA,NE and E in the rats with 60 minute reperfusion after 30 minutes brain ischemia.Results:The ET content in the AMG was higher than that in the ACG.The decrease of ET content in the HYHD,the XXNG,the RG and increase of CGRP in the LYHD were prominent compared with those in the AMG.The excitability of sympathetic-adrenal system was improved in the AMG,and this excitability in every taking medicine group was decreased,espe-cially in the RG and the XXNG.Conclusion:The enhance of sympathetic-adrenal system's excitability and the CGRP-ET dysequilibrium were correlated with brain ischemia reperfusion injury in the aged rats.Every Chinese traditional drugs in this trial could alleviate the gastrointestinal injury via restraining the excitability of sympathetic-adrenal system and regulating the balance between CGRP and ET.
