摘要
目的研究MTT染色法用于检测细胞凋亡的可行性。方法以As2O3诱导K562细胞建立凋亡细胞模型,并比较以下3种方法检测凋亡的结果。MTT染色法检测、直接在光镜下进行形态学观察以及DNA凝胶电泳。结果MTT染色法可准确判定细胞凋亡,并可清晰分辨正常细胞、凋亡细胞和死亡细胞。结论MTT染色法是一种简单、可行的检测细胞凋亡的方法。
Objective To studythefeasibilityof usingMTT assayto detectcellapoptosis.Methods K562cellsweregrown in RPMI1640mediumsupplementedwith10%calfserumand4μmol/Larsenictrioxide.Apoptosiswas inducedin the culturedcellsby As 2 O 3 ,andthecellsweredetectedwithopticalmicroscope,DNAgelelectrophoresisandMTT staining respectively.Result MTT stainingcouldalsoaccuratelydetectcellapoptosis,by whichtheapoptoticcellswereeasily distinguishedfromnormalcellsanddeadcells.Conclusion MTTstainingis simple,convenientandpracticalfordetecting cellapoptosis.
