摘要 目的 研究水溶性药物骨架片释放达零级的方法。方法 以盐酸氢吗啡酮 (hydromorphonehydrochloride)为模型药物 ,用熔融制粒法制备骨架片 ,用HPLC测定骨架片的释放度。结果 通过压制多层片和混合使用骨架材料 ,可解决盐酸氢吗啡酮骨架片前期释放偏快的问题 ,使整个释放近零级。结论 通过处方和工艺调整可以获得具有零级释放特征的水溶性药物骨架片。 OBJECTIVE: To study methods to prepare matrice containing soluble drug with zero-order release kinetics. METHODS: The hydromorphone hydrochloride matrice was produced by melt granulation. The concentrations during the dissolution was determined by HPLC. RESULTS: Through controlling the release area of matrice at the beginning of dissolution, incerasing the hydrophobicity of the matrix surface or shortenning the time to form the diffusion-retarded layer on the matrix surface, zero-order release of soluble drug was attained. CONCLUSION: Through adjusting the formulation and preparation process, the matrice containing soluble drug, whose release behavior fited with zero-order kinetics, can be made.
