摘要
目的建立肉苁蓉一测多评含量测定方法,同时测定管花肉苁蓉和荒漠肉苁蓉药材中6种苯乙醇苷类成分。方法采用HPLC-DAD,以松果菊苷为参照物峰,建立其与毛蕊花糖苷、肉苁蓉苷C、肉苁蓉苷A、2-乙酰毛蕊花糖苷、6-乙酰毛蕊花糖苷的相对校正因子,分别以外标法和一测多评法计算6种苯乙醇苷类成分的含量,比较计算值和外标法实测值之间的差异。结果经过方法学考察和样品测定,肉苁蓉中各成分相对松果菊苷的校正因子分别为f毛蕊花糖苷/f松果菊苷=1.008,f肉苁蓉苷C/f松果菊苷=1.871,f肉苁蓉苷A/f松果菊苷=0·809,f2-乙酰毛蕊花糖苷/f松果菊苷=0.611,f6-乙酰毛蕊花糖苷/f松果菊苷=0.603。在不同实验条件下相对校正因子重复性好,与外标法相比无显著性差异。结论该方法准确、简便,重复性好,能同时对肉苁蓉药材中多种有效成分进行定性定量分析,为肉苁蓉药材的质量控制提供实验方法。
Objective To establish a HPLC method for simultaneous determination of 6compounds including echinacoside, cistanoside C,cistanoside A,acteoside,2'-acetylacteoside and 6'-acetylacteoside in Cistanche tubulosa and Cistanche desertico- la by quantitative analysis of multi -components with a single -marker (QAMS).Methods By using echinacoside as internal reference,the relative correction factor (RCF)of cistanoside C,cistanoside A,acteoside,2'-acetylacteoside and 6'-acetylacte- oside were established,and the above 6phenylethanoid glycosides constituents were determinate and compared by QAMS and standard curve meti/od,respectively.Results The relative correlation factors of acteoside,cistanoside C,cistanoside A,2'- acetylacteoside and 6'-acetylacteoside were calculated by methods validated as 1.008,1.871,0.809,0.611,and 0.603,respectively.The determination results were compared with those data obtained by the standard curve method which reflected that the good reproducibility of the relative correlation factors,and no significant difference in assay results was observed between the external standard method and QAMS method.Conclusion The method is simple and reliable,which can be readily utilized for simultaneous distinguish and determine the content of several bioactive constituents in Cistanche tubulosa and Cistanche deserticola,as well as their quality control.
