二氢杨梅素改善抑郁样行为及应激诱导认知障碍的作用 被引量:4

Dihydromyricetin ameliorates depression-like behaviors and stress-induced cognitive dysfunction

摘要 目的研究二氢杨梅素(DHM)对抑郁样行为及学习记忆的影响。方法将小鼠随机分成无应激+溶媒组(NS+Veh)、无应激+DHM组(NS+DHM)、应激+溶媒组(CRS+Veh)以及应激+DHM组(CRS+DHM),应激组小鼠经历10 d应激,DHM组小鼠在应激后5 d注射DHM,应激结束后所有小鼠依次进行旷场、新颖物体识别、Y迷宫、悬尾、强迫游泳等实验测试,观察DHM对小鼠的自发活动、学习记忆及抑郁样行为的作用。将WKY大鼠随机分为两组Veh组和DHM组,一部分大鼠(每组8只)依次进行旷场和FST测试,另一部分(每组4只)断头取脑,采用Western blot法检测海马内脑源性神经营养因子(BDNF)的水平。结果 CRS+Veh组小鼠在Y迷宫和新颖物体识别测试中的学习记忆能力显著低于NS+Veh组,CRS+DHM组小鼠在Y迷宫和新颖物体识别测试中的学习记忆能力显著高于CRS+Veh组,而与NS+Veh组比较,NS+DHM组在Y迷宫和新颖物体识别测试中的学习记忆能力无显著性差异; CRS+Veh组小鼠在悬尾和强迫游泳测试中的不动时间显著高于NS+Veh组,CRS+DHM组小鼠在悬尾和强迫游泳测试中的不动时间显著低于CRS+Veh组,同时,与NS+Veh组比较,NS+DHM组的悬尾和强迫游泳测试中的不动时间显著降低。在WKY大鼠中,DHM组在强迫游泳测试中的不动时间显著低于Veh组;在旷场测试中两组大鼠穿越的格子数无显著性差异。结论 DHM改善应激诱导小鼠记忆损伤,但对正常小鼠学习记忆无影响; DHM可改善正常小鼠、慢性应激小鼠、WKY大鼠的抑郁样行为以及上调WKY大鼠BDNF的表达。 OBJECTIVE To study the effect of dihydromyricetin(DHM)on depression-like behaviors and memory.METHODS Mice were randomly divided into non -stress +vehicle group (NS +Veh),non -stress +DHM group (NS +DHM),chronic restrain stress +vehicle group (CRS +Veh),and chronic restrain stress +DHM group (CRS +DHM),stress mice were exposed to 10 consecutive days of CRS.DHM or Vehicle were administrated last 5days of CRS.Then,the open -field test(OFT),novel object recognition test(NOR),Y-maze test(YMT),tail suspension test (TST),and forced swim test(FST)were performed to examine the effect of DHM on the spontaneous activity,memory,and depression -like behavior in mice respectively.In addition,WKY rats were randomly divided into Veh group and DHM group.Some of them were subjected to open -field test and forced -swimming test.The others were decapitated and detected for BDNF levels in the hippocampus by Western blot.RESULTS The memory of CRS +Veh mice were significantly decreased in the YMT and NOR than that in the NS +Veh group.The CRS +DHM mice had higher memory in the YMT and NOR than in the CRS +Veh group.There was no significant difference in learning and memory between NS +DHM group and NS +Veh group in YMT and NOR.The immobility time of CRS +Veh mice in TST and FST was significantly higher than that in NS +Veh group.The immobility time of CRS +DHM mice in TST and FST was significantly lower than that of CRS +Veh group.At the same time,the immobility time of the NS +DHM group was significantly higher than that of the NS +Veh group in TST and FST.In WKY rats ,the immobility time of DHM group in FST was significantly lower than that of Veh group.There was no significant difference in the number of crossing between the two groups of rats in the open field test.CONCLUSION DHM can ameliorates the stress - induced memory impairment in mice but not influence the memory in normal mice.In addition,DHM can ameliorates the depression -like behavior of normal mice,CRS mice and WKY rats,and up -regulates BDNF expression in WKY rats.
