摘要
目的 探讨特发性肺纤维化 (IPF)对老年小鼠肠黏膜屏障的影响。方法 16只健康老年小鼠随机分为对照组及IPF组。IPF组气管内注射博莱霉素形成IPF模型,造模后28d处死小鼠,分别留取血清、肺脏及小肠,行酶联免疫吸附试验、苏木精-伊红染色及SABC染色观察肠黏膜屏障损伤的相关指标及病理变化。结果 IPF组小鼠肺部 HE染色提示肺纤维化模型成功。IPF组小鼠血清丙二醛较对照组明显增高,总超氧化物歧化酶、谷胱甘肽过氧化物酶均低于对照组,差异均有统计学意义 (t值分别为3.177、2.770、2.450,P 值均<0.05)。IPF组小肠绒毛水肿,边界不清。绒毛高度较对照组显著降低 (t=5.284,P <0.01)。IPF组紧密连接蛋白闭锁小带蛋白1、闭锁蛋白吸光度值均显著低于对照组 (t值分别为4.631、2.277,P 值均<0.05)。结论 IPF影响老年小鼠肠黏膜绒毛长度及细胞间紧密连接蛋白含量,引起肠黏膜屏障损伤。
Objective To investigate the effect of idiopathic pulmonary fibrosis (IPF)on intestinal mucosal barrier in elder mice.Methods 16 healthy elderly mice were randomly divided into control group and IPF group,IPF group received intratraeheal injection of h[eomyein to form IPF model.After 28 days, all the mice were executed,with the serum,lung and small intestine being collected respectively.Methods of enzyme linked immunosorbent assay,hematoxylin-eosin staining and SABC staining were used to observe the correlation indexes and pathological changes of intestinal mucosal barrier injury.Results The photo of lung HE staining in IPF group suggested IPF model was successful.The figure of malondialdehyde in IPF group was significantly higher than control group,the statistics of total superoxide dismutase and glutathione peroxidase were lower than control group,the differences were statistically significant (t =3.177,2.770,2.450,all P <0.05).Compared with control group,the height of intestinal villi in IPF group is significantly decreased (t =5.284,P <0.01),with the intestinal villi being dropsical and having obscure boundary.Absorbance values of tight junction protein zonula occludens-1 and Occludin in IPF group were significantly lower than control group (t =4.631,2.277,both P <0.05).Conclusions IPF causes intestinal mucosal barrier damage by affecting the height of intestinal mucosal villi and the content of intercellular tight junction proteins in elderly mice.
