青蒿素硫酸羟氯喹复方对大鼠佐剂性关节炎的作用研究 被引量:7

摘要 目的观察青蒿素硫酸羟氯喹复方(Artemisinin-Hydroxychloroquine,AH)对佐剂性关节炎模型大鼠的作用,探讨AH治疗类风湿性关节炎(Rheumatoid Arthritis,RA)的药效及机制。方法于大鼠右后足跖皮内注射Freund’s完全佐剂致炎造模,成模后连续给药至致炎后第30天,并记录实验期间大鼠的体质量、足肿胀度、关节炎指数;给药结束后,采集腹主动脉血清、左足踝关节组织;应用酶联免疫吸附法检测大鼠血清中干扰素γ(IFN-γ)的水平,并进行踝关节组织病理学检查,综合评价药物对大鼠佐剂性关节炎的影响。结果与模型对照组比较,AH的中、高剂量组明显降低AA大鼠继发性足肿胀度(P<0.05),并且减轻踝关节的破坏程度,AH的低、中、高剂量组均明显降低关节炎指数(P<0.05)和炎性因子IFN-γ水平(P<0.01)。结论 AH的中、高剂量组对大鼠佐剂性关节炎有一定的抗炎作用,可缓解佐剂性关节炎大鼠的关节炎症状,并且可能会缓解、修复关节病理损伤。 Objective To observe the effect of artemisinin-hydroxychloroquine (AH) on adjuvant arthritis model rats, and to explore the efficacy and mechanism of AH in the treatment of rheumatoid arthritis (RA). Methods Freund's complete adjuvant was injected into the right hind paws of rats to induce inflammation. The model rats were administered continuously until the 30th day after inflammation. During the experiment, the weight, paw swelling and arthritis index of the rats were recorded. At the end of experiment,rats were sacrificed and the serum and left ankle were collected;the concentration of IFN-γ in the serum was detected by enzyme-linked immunosorbent assay (ELISA),and the histopathological examination of the ankle joint was performed to evaluate the effect of the drug thoroughly. Results Compared with the control group,the medium and high dose groups of AH significantly reduced the paw swelling of adjuvant arthritis (AA) rats (P < 0.05) and alleviated the destruction of the ankle joint. The low, medium and high dose groups of AH significantly reduced both the arthritis index (P < 0.05) and IFN-γ concentration (P < 0.01). Conclusion The medium and high dose groups of AH could alleviate the arthritis symptoms and have certain anti-inflammatory effects on AA rats,and might alleviate and repair the damage of joints.
