高效液相色谱法测定黑果腺肋花楸提取物中氯化矢车菊素的含量及抗氧化性研究 被引量:7

摘要 建立黑果腺肋花楸提取物中氯化矢车菊素的含量测定方法,并优化黑果腺肋花楸提取物供试品的制备方法。以氯化矢车菊素的含量为指标,采用高效液相色谱法测定黑果腺肋花楸提取物中氯化矢车菊素的含量,色谱柱:伊利特ODS2 C18(4.6 mm×250 mm,5μm);流动相以甲醇-0.2%磷酸水溶液(v/v);进样量20μL;检测波长276 nm;柱温30℃。在建立含量测定方法的同时探索黑果腺肋花楸提取物水解提取氯化矢车菊素的工艺条件。试验结果表明氯化矢车菊素在3.6~36.0μg.mL^(-1)(r=0.9995)线性关系良好;氯化矢车菊素的最佳水解提取工艺为:水解温度100℃、水解时间2 h、溶剂酸浓度3%盐酸甲醇。在优化水解提取条件下,氯化矢车菊素的含量为1.8570 mg.g^(-1)。该含量测定方法具有良好的精密度,结果准确可靠;优化的提取方法快速、简单,为黑果腺肋花楸果的开发利用提供了依据。体外抗氧化活性的试验结果表明,黑果腺肋花楸提取物对DPPH自由基具有清除活性,且随着其质量浓度的增加,清除活性明显增强;利用FRAP法和钼酸铵法测得黑果腺肋花楸提取物具有一定的抗氧化活性。 To establish a method for the determination of cyanidin chloride in the extract ofAroni amelanocarpa,and to optimize preparation method of Aroni amelanocarpa extract,cyanidin chloride content was selected as an index, cyanidin chloride content in Aroni amelanocarpa extract was determined by high performance liquid chromatography (HPLC).The chromatographic separation was performed in Illiot ODS2 colum (C18,4.6 mm × 250 mm ,5 μm). The mobile phase was eluted with carbinol-0.2%ortho-phosphoric acid.The injection volume was 20 μL and the detection wave length was 276 nm.The column temperature was 30 ℃ .The technological conditions for the hydrolysis extraction of cyanidin chloride from Aroni amelanocarpa were explored while establishing a determination method. The test results showed a good linear relationship of cyanidin chloride at 3.6 -36.0 μg.mL-1 (r=-0.9995).The best hydrolysis extraction process of cyanidin chloride were:the hydrolysis temperature was 100 ℃,the hydrolysis time was 120 min and the solvent acid concentration was 3% carbinol hydrochloric acid.The content of cyanidin chloride was 1.8570 mg·g^-1 under the optimized hydrolysis extraction conditions.The method of determination has good precision and the result is accurate and reliable.The optimized extraction method is fast and simple which provides the basis for the development and utilization of the Aroni amelanocarpa.The results of antioxidant activity in vitro showed that,the extract of Aroni amelanocarpa has scavenging activity for DPPH free radical,and the scavenging activity increased with the increase of mass concentration;frap and ammonium molybdate methods were used to determine the antioxidant activity ofAroni amelanocarpa extract.
