木薯微茎尖离体脱毒与病毒检测技术分析

Study on Virus-free Culture and Viral Detection Techniques on Cassava(Manihot esculenta Cranz) Micro-shoot Tip in Vitro

摘要 为研究木薯微茎尖离体脱毒和病毒检测技术体系建立的条件,本研究以感病‘华南8号’('SC8')微茎尖为外植体,比较了茎尖大小、病毒唑浓度对‘SC8’微茎尖脱毒效果的影响,用RT-PCR方法进行病毒检测。结果表明,微茎尖大小为0.4~0.5 mm在MS+6-BA 0.01 mg/L+NAA 0.02 mg/L+GA31.0 mg/L培养基上,成活率为61.7%,脱毒率为35.1%;培养基中附加10 mg/L病毒唑微茎尖的成活率略微下降,为57.7%,脱毒率提高至42.1%。本研究建立了SC8木薯微茎尖离体脱毒培养与病毒检测技术体系,为木薯健康种苗生产和国家与地区之间安全交流木薯种质资源提供技术参考。 In order to establish virus-free culture in vitro and the detection techniques on cassava(Manihot esculenta Cranz) micro-shoot tip system. In this study, the infected virus micro-shoot tips of cassava cultivar ’Huanan8’(’SC8’) used as explant. The effects of micro-shoot tip sizes, ribavirin concentration were investigated by comparison test, and the virus was detected with RT-PCR. The results showed that the size of the micro-shoot tip is the main factor both on survival and virus-free rate;the survival rate of the size 0.4~0.5 mm cassava micro-shoot tip was 61.7%, cultured in MS+0.01 mg/L 6-BA+0.02 mg/L NAA+1.0 mg/L GA3 medium, and the virus-free seeding rate was 35.1%. The survival rate was slightly declined to 57.7% with addition of 10 mg/L ribavirin to the culture medium, and the virus-free seeding rate was increasing to 42.1%. The virus-free culture in vitro and the viral detection techniques of micro-shoot tip culture on cassava SC8 cultivars were successfully establishment which could provide the technical support for cassava seedling healthy production and the security exchange of cassava germplasm between countries and regions.
