摘要
目的 通过药效学研究 ,确定与北苍术临床功效、主治基本吻合的有效成分 β 桉叶醇 ,以便为中药材苍术质量标准规范化研究提供药效学实验依据。方法 以正常、新斯的明负荷及脾虚小鼠为实验研究模型 ,观察了茅苍术的有效成分 β 桉叶醇、茅术醇 +β 桉叶醇混合物对小鼠胃肠推进功能及脾虚模型小鼠体征、体重、胃肠推进功能的影响。 结果 β 桉叶醇 1次性口服给药可显著促进正常小鼠的胃肠运动 (P <0 .0 5 ,P <0 .0 0 1) ,且呈现出很好的量效关系 ;同时也可证明茅术醇 +β 桉叶醇混合物在剂量与 β 桉叶醇相同的条件下 ,其作用强度与单用 β 桉叶醇无显著差别 ;β 桉叶醇对新斯的明负荷小鼠引起的胃肠运动加快有明显的拮抗作用 (P <0 .0 5 ,P <0 .0 0 1) ;β 桉叶醇可明显改善脾虚模型小鼠的体征 ,使脾虚模型小鼠的体重上升 ,显著抑制脾虚小鼠的胃肠运动 (P <0 .0 5 ,P <0 .0 1) ,使脾虚小鼠的胃肠运动趋于正常。结论 β 桉叶醇可以作为苍术健脾燥湿作用的有效活性成分之一。茅术醇 +β
OBJECTIVE To search for the active constituents contributed to the underlying therapeutic effects and clinical indications of Atractylodes chinensis by a pharmacodynamics-oriented chemical isolation and analysis, and provide a pharmacodynamic basis for standardizing the quality control of the herb.METHODS The effects of β-eudesmol,or its combination with hinesol on gastrointestinal peristalsis was investigated in healthy mice, neostigmine-treated mice and mice with spleen deficiency,respectively.RESULTS Oral administration of β-eudesmol at a single dose was found to significantly promote gastrointestinal movement in a dose-dependent manner in healthy mice.(P<0.05),and the effect was stronger when a combination of β-eudesmol and hinesol was used. A remarkable antagonistic effect of β-eudesmol against the increase of gastrointestinal movement induced by neostigmine was observed in mice (P< 0.05).An improvement such as the increase of body weight,the normalization of gastrointestinal movement was observed after treatment with β-eudesmol in spleen-deficient mice.CONCLUSION β-eudesmol and hinesol can be considered as the active constituents for invigorating spleen to eliminate dampness in Rhizoma Atractylodis.
