全波长扫描对油菜籽粒中硫代葡萄糖甙的测定 被引量:3

Determination of Glucosinolates Content in Seeds of Rapeseed by Full Spectrum Scan

摘要 首次利用全谱多组分分析方法对油菜籽粒中硫代葡萄糖甙 (下称硫甙 )进行了测定 ,利用紫外 /可见光分光光度计可将甘蓝油菜 6种主要硫甙 (PRO、GNL、GNA、GBN、4-OHGBS、GBS)同时检测出来。在波长为 2 1 0~ 40 0 nm范围内 ,对样品进行全波长扫描 ,扫描数据经傅里叶变换 ,采用目标因子分析法 (TFA)和最小二乘 (LS)回归分析法进行多变量统计分析。首先以 2 0份标准样品建立数学校正矩阵 ,并对其进行了优化 ,优化的操作参数值为 :启始项 =0、项数目 =1 6、矢量数目 =8,优化后的数学校正矩阵标样的总 SEE=1 .3 2 0、% Resid=0 .1 0 8,然后对样品进行了分析 ,样品的 % Resid=0 .1 3 8、CV≤ 5 .92 %。与 HPLC测定法比较 ,结果准确 ,而样品不用分离 ,操作简便 ,速度快 ,重复性好 。 At first time this paper showed full spectrum multicomponent analysis method to determine glucosinolates content of rapeseed.Six main components(PRO,GNL,GNA,GBN,4 OHGBS,GBS) of glucosinolates contents in rapeseed were determined simultaneously with UV/VIS spectrophotometer.To make a full spectrum scan to samples in 210 to 400 nm wavelength.The scan data with fourier transform was made multi variable statistical analysis through target factor analysis and least squares regression analysis.In the experiment, a calibration matrix first was calculated based on 20 standard samples, and the matrix was optimized.Optimum operational parameters were: starting term was 0,number of terms was 16, and number of vectors was 8. Statistical information of standards of the optimum calibration matrix was:total SEE was 1.320,% Resid was 0.108.Then, the samples analysis was performed.The results of analysis were: %Resid was 0.138, CV ≤5.92%. The approach had similar result to HPLC, and the sample didn′t need to separate,it was operation was simpler and quicker than HPLC, and it was accuracy and repeatability were well. The method had a better utilization value.
