摘要
目的 考察甘草和五味子对大鼠肝微粒体CYP45 0的诱导作用及其对丙咪嗪体外代谢的影响。方法 大鼠分别给予甘草或五味子的水提物连续 3d或 6d ,然后制备肝微粒体并测定其CYP45 0的水平 ;以丙咪嗪为底物 ,采用体外温孵法 ,用HPLC测定丙咪嗪及其主要代谢物的量以考察其代谢情况。结果 给予中草药的大鼠肝微粒体CYP45 0水平均显著提高 ,其对丙咪嗪的体外代谢速率显著加快。结论 研究显示甘草和五味子对大鼠CYP45 0有诱导作用 。
OBJECTIVE To observe the alteration of the hepatic CYP450 content and imipramine metabolism in rats after oral administration of the traditional Chinese medicines (TCM), Glycyrrhiza uralensis Fisch and Schisandra chinensis Baill.METHOD The rats were given orally the aqueous extracts of the two kinds of TCMs for three or six days. Then the liver microsomes were prepared and the P450 level were determined. Imipramine was incubated with the liver microsomes, and its metabolism was estimated by measuring the concentration of the parent drug and its metabolites in the incubation mixture by means of HPLC.RESULTS The cytochrome P450 content and the metabolism rate of imipramine were enhanced significantly in the liver microsomes from the rats pretreated with the two TCMs.CONCLUSION Glycyrrhiza uralensis Fisch and Schisandra chinensis Baill showed induction effect on hepatic CYP450 level in rats in a dose and/or time dependent manner.
