摘要
血脑屏障的存在使大分子药物难以进入脑中发挥疗效,成为中枢神经系统疾病治疗的瓶颈。本文就血脑屏障的结构特点、大分子药物转运入脑的途径及药物与载体间的连接策略等问题进行了综述。
Delivery of macromolecular therapeutics to the brain is severely hindered by the brain capillary endothelial wall which makes up the blood-brain barrier(BBB)in vivo,it has already become a problem in the treatment of the central nervous system disorders.Strategies for the drug transport to brain include:①neurosurgical-based(intra-ventricular drug infusion)②hyperosmotic opening the BBB③brain transport vectors(receptor-mediated vectors and absorptive-mediated vectors).This paper summarized the constructive characteristic of BBB,the transport mechanism of drug across the BBB and the ligation strategy between drugs and vectors.In conclusion,the study of neurophar-maceuticals in the future will require the development of both the drug discovery and the drug delivery.
