摘要
目的:通过去氢骆驼蓬碱 (HM)微球与羟基喜树碱 (HCPT)注射液对大鼠正常肝细胞 (BRL )的毒性作用的比较研究 ,为 HM的临床应用提供佐证。方法:采用 MTT法测试一定浓度范围内 HM与 HCPT对大鼠正常肝细胞 (BRL)的细胞毒作用大小。 结果 :在一定浓度范围内 ,HM与 HCPT的细胞毒作用与药物浓度均呈线性关系 ;针对 BRL 细胞 ,在一定的浓度范围内 HM的细胞毒作用较 HCPT注射液的细胞毒作用为小 ,经检验差别有统计学意义 (P <0 .0 1)。结论:MTT法操作简单、快速、敏感 ,是较好的体外实验方法之一。HM微球新剂型对 BRL的毒性作用小于 HCPT注射液 ,体外实验表明其毒副作用小 ,结合其剂型的靶向性、缓释性、栓塞性等特点 。
Objective: In order to offer evidence for the clinical application of HM(harmine), toxicity of HM and HCPT(hydroxycamptothecin) against the rat normal liver cell (BRL) was compared. Methods: Using the MTT(3 (4,5 dimethylthiazol 2 yl) 2,5 diphenyl tetrazolium bromide) method to detect the cell toxic effect of HM and HCPT against the rat normal liver cell (BRL) at certain concentration scope. Result: There was line relationship between the cell toxicity of HM and HCPT and their drug concentrations; the toxicity of HM against the BRL cell was smaller than the HCPT with statistical significance (P<0.01) . Conclusions: MTT method is a simple\, rapid and sensitive method for assessment of cell toxicity. HM micro spheres is a kind of new drug formulation of HM, its cell toxic effect against BRL is smaller than HCPT injection. In vitro, the low toxicity of the HM micro spheres is found, combing its other characters, i.e, targeting\, slow releasing and embolism, there may have a potential clinical application for the HM micro spheres against the liver cancer and alveolar echinococcosis.
