摘要
目的:研究半乳糖凝集素-3(Galectin-3)在大肠癌中的表达,并探讨其在大肠癌进展过程中的意义,旨在为临床诊断、治疗、预后提供新的理论依据。方法:应用免疫组化法检测52例大肠癌组织、20例大肠腺瘤和20例大肠正常粘膜组织中Galectin-3的表达情况,分析其表达结果与患者的性别、年龄、肿瘤发生部位、肿瘤组织分化程度、浸润程度及淋巴结转移之间关系。实验资料的统计学处理通过SPSS13.0统计软件完成,等级资料采用秩和检验,以P<0.05为差异有统计学意义。结果:①Galectin-3在大肠正常黏膜、大肠腺瘤和大肠癌中均有阳性表达,表达强度依次增加,三组之间阳性表达率差异有统计学意义(P<0.01)。②Galectin-3在浸润程度深的大肠癌组织的表达明显高于浸润程度浅的(P<0.01);Galectin-3在低分化大肠癌中的表达则高于高分化和中等分化的(P<0.05);Galectin-3有淋巴结转移的大肠癌组织中的表达明显高于无淋巴结转移的(P<0.01)。Galectin-3在不同性别、年龄(≤60,>60)及肿瘤发生部位的表达则无明显差异(P>0.05)。结论:大肠癌组织中Galectin-3的表达水平明显高于正常大肠黏膜组织,提示Galectin-3的高表达在大肠癌的发生、发展过程中起到了重要的作用。Galectin-3在大肠癌不同浸润深度、不同分化程度及有无淋巴结转移中的表达有显著差异,提示上述指标在大肠癌的浸润、生长及远处播散过程中起到了一定的作用,并可作为判断大肠癌转移及预后的预测因素。
Objective：To study the expression of Galectin-3 in colorectal carcinoma , and to investigate its in colorectal cancer progression of meaning in the course , designed for clinical diagnosis , treatment , prog-nosis and provide a new theoretical basis .Method： Detected the expression of Galectin-3 in 52 cases of colorectal cancer , 20 cases of colorectal adenoma and 20 cases of colorectal normal mucosa tissue through the application of immunohistochemistry .Analyzed the relationship between its expression with the patient ’s sex, age, tumor location, tumor differentiation, infiltration and lymph node metastasi .Result： ①Galectin-3 in colorectal normal mucosa , colorectal adenoma and carcinoma showed positive expression , and the expression intensity increases , positive expression rate had a significant difference （ P〈0.01） among three groups .②Ga-lectin-3 in infiltration over the muscle layer of the colorectal carcinoma was higher than that of infiltration does not exceed the muscular layer of the colon cancer tissue （ P〈0.01）;Galectin-3 in poorly differentiated colorectal carcinoma was higher than that of well-differentiated and moderately differentiated colorectal canc-er tissue （ P〈0.05 ）; Galetctin-3 in lymph node metastasis of colorectal cancer tissues than those without lymph node metastasis in colorectal cancer tissue （P〈0.01）.Conclusion： The expression of Galectin-3 in colorectal carcinoma is higher than that in normal colorectal mucosa .It suggests that the higher expression of Galectin-3 play an improtant role in the development of colorectal carcinoma .The expressions of Galectin-3 is significantly correlated with depth of invasion , lymph node metastasis and degree of differentiation in color-ectal carcinoma .So they could be a predictive and prognostic factor of colorectal carcinoma .
