摘要
目的通过观察人参益智口服液对樟柳碱所致记忆障碍模型小鼠记忆能力和血清酶学的影响,探讨人参益智口服液改善记忆功能。方法将C57/BL雄性小鼠随机分为6组:空白对照组、模型组、阳性药对照组和人参益智口服液高、中、低剂量治疗组,采取樟柳碱复制小鼠记忆障碍模型。空白对照组给予生理盐水,阳性药对照组给予吡拉西坦,治疗组分别给予高、中、低剂量人参益智口服液,连续给药30 d。采用行为学实验包括Morris水迷宫、跳台和避暗实验法检测小鼠记忆能力,并检测血清SOD、MDA的变化。结果 Morris水迷宫检测结果显示,与对照组相比,模型组小鼠平均潜伏期延长及平均跨越平台次数明显减少,说明樟柳碱能够造成学习记忆障碍模型。人参益智口服液高、中剂量组能够减少潜伏期时间和增加平均跨越平台次数(P<0.05)。跳台和避暗实验结果显示,与模型组相比,人参益智口服高、中剂量组平均潜伏期延长,平均错误次数减少(P<0.05,P<0.01)。血清酶学检测显示人参益智口服液能够增加血清SOD,降低MDA水平(P<0.05,P<0.01)。结论人参益智口服液能够改善樟柳碱造成的小鼠学习记忆障碍。
Objective To observe the effect of Ginseng Yizhi Oral Liquid on memory ability and serum enzymes induced by anisodine in mice and to explore the function of Ginseng Yizhi Oral Liquid. Methods C57/BL male mice were randomly divided into 6 groups：blank control group, model group, positive drug control group, high, medium, low, dose treatment groups. A model of memory impairment was established by anisodine. The blank control group was given physiological saline. The positive drug control group was given piracetam. The treatment group was given high, medium, low dose of Ginseng Yizhi Oral Liquid for 30 days. The memory abilities of mice were observed by using step-through test, step-down test and Morris water maze. The content of SOD and MDA in blood serm of C57 mice was detected. Results Morris water maze showed mean incubation period was prolonged and the average number of crossing platform was reduced in the model group compared with the control group. Therefore, anisodine can establish model of learning and memory disorder. The mean incubation peri- od was reduced and the average number of crossing platform increased in high, medium, low, dose treatment groups （ P 〈 0.05）. According to the experiments, the mean incubation periods in high, medium, low, dose treatment groups were prolonged compared with that of the model group（ P 〈 0.05, P 〈 0.01 ）. Detection of serum enzymology showed that Ginseng Yizhi Oral Liquid can increase senma SOD and reduce the level of MDA （ P 〈 0.05, P 〈 0.01 ）. Conclusion Ginseng Yizhi Oral Liquid can improve learning and memory abilities of mice with learning and memory disorders induced by aniso- dine.
