摘要
目的观察佛司可林(forskolin,FSK)、异佛司可林(isoforskolin,ISOF)对SD大鼠的心血管系统、呼吸系统的影响.方法 (1)将50只SD大鼠按雌雄各半随机分为5个处理组,即对照组(Control)、FSK及ISOF低、高剂量组(2、6 mg/kg),每组n=10.(2)动物颈总动脉置管,连接压力换能器及多道生理记录仪,记录大鼠心率(heart rate,HR)、呼吸频率(breathing rate,BR)、收缩压(systolic blood pressure,SBP)、平均动脉压(mean arterypressure,MAP)、舒张压(diastolic blood pressure,DBP).待各监测指标平稳后给予相应记录,十二指肠给药,分别记录大鼠给药前、给药后0.5 h、1 h、1.5 h、2 h和2.5 h的心率、呼吸频率、收缩压、平均动脉压、舒张压情况.结果与给药前比较,各处理组在给药后0.5 h、1 h、1.5 h、2 h和2.5 h的心率、呼吸频率、收缩压、平均动脉压、舒张压均无明显变化,各处理组均差异无统计学意义(P>0.05);与对照组比较,各处理组在给药前、给药后0.5 h、1 h、1.5 h、2 h和2.5 h的心率、呼吸频率、收缩压、平均动脉压、舒张压均差异无统计学意义(P>0.05),各处理组差异无统计学意义(P>0.05).结论一般药理学研究表明,FSK及ISOF低、高剂量组(2、6mg/kg)对SD大鼠的心率、呼吸频率、收缩压、平均动脉压、舒张压均无明显影响.
Objective To investigate whether Forskolin (FSK) , Isoforskolin (ISOF) has any influences on cardiovascular system and respiratory system of SD rats. Methods (1) 50 SD rats were divided into five groups randomly: Control group (Control), the low (2 mg/kg) and high (6 mg/kg) dose FSK and ISOF group, n = lO in each group, with the proportion of male and female 1:1. 2) Carotid artery catheters were used for those SD rats, pressure transducer and the multi-channel physiology recorder were applied to record in the heart rate, breathing frequency, systolic blood pressure, mean arterial pressure, diastolic blood pressure. The data were recorded after the monitoring index got stable. Medicines were given from the duodenum after the rats were anesthetized, the heart rate, breathing, systolic blood pressure, mean arterial pressure, diastolic blood pressure were recorded before and 0.5 h, 1 h, 1.5 h, 2 h and 2.5 h after the drug administration in treatment group. Results Compared with before drug administration, the heart rate, breathing rate, systolic blood pressure, mean arterial pressure, diastolic blood pressure of the treatment group had no significant difference 0.5 h, 1 h, 1.5 h, 2 h and 2.5 h after drug administration. Compared with control group, the heart rate, breathing, systolic blood pressure, mean arterial pressure, diastolic blood pressure had no significant difference between before and 0.5 h, 1 h, 1.5 h, 2 h and 2.5 h after drug administration. Conclusion General pharmacology study shows that the low (2 mg/kg) and high (6 mg/kg) dose of FSK and ISOF has no significant effect on heart rate, breathing, systolic blood pressure, mean arterial pressure, diastolic blood pressure of SD rats.
