摘要
目的探究海地瓜岩藻聚糖硫酸酯(Acaudina molpadioides fucoidan,Am-FUC)对高脂高糖饲料诱导的2型糖尿病小鼠肾脏的保护作用。方法采用饲喂高脂高糖饲料的方法建立2型糖尿病小鼠模型,各组小鼠饲喂相对应的饲料19 w后,分别检测小鼠空腹血糖、口服葡萄糖耐量、血清胰岛素、胰岛素抵抗指数,尿液中尿素氮(UN)、肌酐(Cr)、尿酸(UA)、尿糖含量,尿液中尿总蛋白、微量白蛋白(mAlb)、β-N-乙酰氨基葡萄糖苷酶(NAG)排泄率,光镜下观察小鼠肾组织的显微结构变化。结果 Am-FUC能降低2型糖尿病小鼠空腹血糖(P<0.05)、口服葡萄糖耐量(P<0.05)、血清胰岛素(P<0.05)、胰岛素抵抗指数(P<0.05)、UN(P<0.01)、Cr、UA和尿糖(P<0.01)含量,显著改善尿总蛋白、mAlb、β-NAG排泄率(P<0.01),有效修复肾小囊肿胀、抑制肾小球肥大、改善肾小管组织空泡化。结论 Am-FUC对2型糖尿病小鼠肾脏具有较好的保护作用。
Objective To study the protective effects ofAcaudina molpadioides fucoidan (Am-FUC) on kidney injury in type 2 diabetic mice fed a high-fat and high-sucrose diet (HFSD). Method Type 2 diabetic model mice were established by feeding HFSD. After 19 weeks, the contents of fasting plasma glucose, oral glucose tolerance, serum insulin and homeostasis model assessment indexs were determined. The levels of urea nitrogen (UN), creatinine (Cr), uric acid (UA) and urine glucose in urine were also detected. The excretion rates of total protein, microalbuminuria (mAlb) and β-N-acetyl-glucosaminidase (NAG) in urine were measured. Microstructure of the kidneys was observed. Result Compared with the type 2 diabetic model group, fasting plasma glucose (P〈0.05), oral glucose tolerance (P〈0.05), serum insulin (P〈0.05), homeostasis model assessment indexs (P〈0.05), and UN (P〈0.01), Cr, UA, glucose (P〈0.01) in urine were remarkably decreased in mice fed Am-FUC. The excretion rates of the total protein (P〈0.01), mAlb (P〈0.01) and NAG (P〈0.01) in urine were sigllificantly decreased. The microstructure of the kidneys was also improved by Am-FUC compared to HFSD-fed mice. Conclusion Am-FUC had markedly protective effects on kidney injury induced by type 2 diabetes.
