摘要
目的:观察针刺百会、内关穴对大脑中动脉阻塞(MCAO)大鼠未折叠蛋白反应(UPR)通路eIF2α因子基因表达的影响,探讨针刺抑制细胞凋亡、保护脑神经元的内在机制。方法:将SD大鼠60只随机分为捆绑组、假手术组、模型组、针刺组、依达拉奉组,每组12只。线栓法制备MCAO大鼠脑缺血再灌注模型,RT-PCR法检测缺血侧顶叶大脑皮层eIF2α基因表达变化。结果:与捆绑组和假手术组比较,模型组、针刺组及依达拉奉组eIF2α表达明显增高(P<0.01或P<0.05);与模型组比较,针刺组及依达拉奉组eIF2α基因表达显著降低(P<0.01);与依达拉奉组比较,针刺组eIF2α表达变化差异无统计学意义(P>0.05)。结论:针刺可以下调脑缺血大鼠未折叠蛋白反应通路eIF2α因子基因的表达,这可能是针刺发挥脑保护作用的内在机制之一。
Objective:To explore the protection mechanism of the acupuncture on transient MCAO rat mode through investigating the effects of the acupuncture on eIF2α genes expression in Unfold Protein Response(UPR) signal pathway. Methods:The 60 rats were randomly divided into bundle group, sham-operation group,model group,acupuncture group, and annaoping-chong pill group, 12 in each. Silk was used for ligation of the middle cerebral artery of rats, to establish MCAO rats mode. Methods of reverse transcription-polymerase chain reaction (RT-PCR) were used to measure the expression of eIF2α mRNA. Results: Compared with the bundle group and sham-operation group, the expression of eIF2α were up-regulated in peri-infarction after cerebral ischemia/reperfusion in model group,acupuncture group and annaoping-chong pill group (P〈0.01 or P〈0.05).The expression level of eIF2α mRNA in intervention models were reduced accordingly (P〈0.01). Compared with the model group,the expression of eIF2α were down-regulated in peri-infarction after cerebral ischemia/reperfusion in acupuncture group and annaoping -chong pill group (P〈0.01 or P〈0.05); Compared with the annaoping-chong pill group, the changes of expression level of eIF2α mRNA in acupuncture group were not statistically significant (P〉0.05). Conclusion:Acupuncture can down-regulated the expression of eIF2α genes in Unfold Protein Response(UPR) signal pathway of MCAO rat mode, and this may be one of the protection mechanism of acupuncture treatment.
