核转录因子-κB在动脉粥样硬化缺血性脑卒中模型大鼠脑组织中的表达 被引量:2

摘要 目的研究核转录因子-κB(NF-κB)在动脉粥样硬化(AS)缺血性脑卒中(CIS)模型大鼠脑组织中的表达规律。方法 48只雄性Wistar大鼠随机分为对照组(8只)和模型组(40只),后者制备动脉粥样硬化大鼠模型,然后再被分为动脉粥样硬化模型假手术组,动脉粥样硬化模型持续缺血0.5h、6h、12h、24h组,上述各组大鼠均为8只。免疫组织化学法检测核转录因子-κB在脑组织中的表达及变化。结果各动脉粥样硬化模型持续缺血组与对照组比较NF-κB表达有明显升高(P<0.05),各动脉粥样硬化模型持续缺血组与假手术组比NF-κB表达也有明显升高(P<0.05),并随时间发生规律性变化了,其中NF-κB在持续缺血6h组达到高峰。结论 NF-κB在动脉粥样硬化基础上出现缺血性脑卒中后引起的脑损伤过程中发挥了重要作用。 Objective To research the expressing rule of nuclear factor κB (NF κB)in the brain tissue of rats model with atherosclerot-ic ischemic brain stroke.Methods Forty eight male Wistar rats were divided into control group (n=8),and experiment group (n=40).Rats of the experiment group were designed to rats model with AS,and then were divided into the sham operation group (n=8). 0.5 h model subgroup,6 h model subgroup,12 h model subgroup,24h model subgroup.The expression of NF κB in the brain tissue was detected by immunohistochemical assay.Results The expression of NF κB in each experiment group was higher than the con-trol group (P<0.05),that in continuous ischemia of the model subgroups was higher than sham operation group (P<0.05).The expression of NF κB reached its peak at the 6h.Conclusion NF κB played an important role in the ischemic brain stroke caused by atheroscle-rosis.
