IL-22诱导小鼠输卵管上皮细胞表达β防御素-2的初步研究 被引量:1

摘要 目的:探讨细胞因子白介素22(IL-22)对小鼠输卵管上皮细胞2型鼠β防御素(mBD-2)表达的影响。方法:流式细胞术检测体外培养的小鼠输卵管上皮细胞膜表面IL-22R表达水平。100ng/ml重组鼠IL-22刺激小鼠输卵管上皮细胞,ELISA检测不同时间(0、12、24、48、72h)mBD-2的表达情况;不同浓度重组人IL-22(0、25、50、100、200ng/ml)刺激小鼠输卵管上皮细胞48h,ELISA检测mBD-2表达情况。结果:小鼠输卵管上皮细胞膜表面表达IL-22R。IL-22诱导小鼠输卵管上皮细胞生成并释放mBD-2表达,具有时间与剂量依赖性。100ng/ml IL-22为最佳干预浓度,且mBD-2表达与IL-22的浓度具有依赖关系。结论:IL-22能显著诱导小鼠输卵管上皮细胞生成并释放mBD-2,且具有时间和剂量依赖性。 Objective:To investigate the effects of IL-22 on the release of murin β-de-fensin-2 in murine oviduct epithelial cell. Methods:The expression of IL-22R on murine ovi-duct epithelial cells using flow cytometry. Cells were stimulated with recombinant mouse IL-22 (rmIL-22) for 0,12,24,48,72 hours. The cells were stimulated with different concentration (0,25,50,100,200 ng/ml) of rmIL-22 for optimal dose. The expression of mBD-2 was detec-ted by ELISA. Results:The membranes of murine oviduct epithelial cells highly expressed IL-22R. 48 hours was an effective time for mBD-2 expression with rmIL-22 treatment,and the optimal concentration was 100ng/ml. Conclusion:The murine oviduct epithelial cells highly ex-press IL-22R,and the interaction of rmIL-22 and IL-22R induces the mBD-2 expression in mu-rine oviduct epithelial cells in a time-and concentration-dependent manner.
