一株高产纤维素酶菌的筛选及发酵条件优化 被引量:2

Optimizing Conditions of Screening and Fermentation for a Strain of High-yield Cellulase

摘要 采用羧甲基纤维素钠(CMC)-刚果红平板法,对富含腐烂玉米秸秆的土样进行纤维素酶产生菌的筛选及目标菌株发酵培养条件的优化试验。结果表明,筛选分离出10株产纤维素酶菌株,其中有3株菌株产酶效果较好,特别是菌株tg31对玉米秸秆水解率高达37.62%,其最佳发酵工艺的温度为28℃,发酵初始pH 5.0,接种量6%,摇瓶转速180 r/min,经5 L罐放大试验,得出在120 h时其滤纸酶活力(FPA)可达到14.21 IU/mL。 To obtain the high-yield cellulase strain,the sodium carboxymethyl cellulose (CMC)-congored plate method was used to screen cellulase-producing bacteria in soil samples riched with corn stalk fiber.Fermentation conditions were optimized also determined.The results showed that 10 cellulase-producing bacterial strains were isolated.Among these strains,three bacterial strains had the best effects on producing the celluloses.Especially,the strain tg31 could hydrolyze corn straw at a rate up to 37.62%.The optimal fermentation conditions were temperature of 28 ℃,initial fermentation with pH 5.0,6% inoculation and shaking speed of 180 r/min.Under the optimal condition,the strain was amplified with 5 L tank and enzyme activity reached 14.21 IU/mL at 120 h.The strain Tg31 obtained the best FPA activity with significant hydrolysis rate of corn stalk,indicating that it had the abroad prospect of application.
