摘要
[目的]提高番茄枯萎病菌拮抗细菌解淀粉芽孢杆菌(Bacillus amyloliauefaciens)S13的芽孢数量和芽孢产率.[方法]采用单因素和正交试验,研究不同的氮源、碳源、pH、温度、培养时间对解淀粉芽孢杆菌形成芽孢的影响.[结果]确定解淀粉芽孢杆菌产芽孢培养基的最佳组分为(g/L):玉米粉0.4,酵母膏0.1,玉米浆1.25,KH2P040.1,K2 HP04 0.1,MgS04·7H20 0.05.对发酵条件进行了研究,产芽孢最适pH为7.0,最适温度为35℃,最适时间为26 h.[结论]通过优化培养基组分和培养基条件,1 000 L罐发酵液中芽孢数达到4.9×10^9cfu/mL,芽孢产率为90%,为进一步该菌株在复合生物肥中的应用打下基础.
[Objective]In order to improve the spore production rate and spore quantity of antagonistic strain S13 against Fusarium wilt.[Method] The medium composition for the production of Bacillus amyloliauefaciens S13 spores was optinized by using the single factor and orthogonal tests.Various factors,such as carbon sources,nitrogen sources,initial pH,temperature and fermentation time were investigated.[Result] The results showed that the optimum medium was as follows (g/L):corn flour 0.4,yeast extract 0.1,corn steep liquor 1.25,KH2PO4 0.1,K2HPO4 0.1,MgSO4 · 7H2O 0.05.The optimum culture conditions were as follows:pH 7.0,temperature 35C and fermentation time 26 h.[Conclusion] Under this optimum condition,the spore quantity of strain S13 reached 4.9 × 10^9cfu/mL and the spore production rate was 90% in fermentation liquid 1 000 L fermentation tank.It laid the foundation for the application of Bacillus velezensis S13 as a kind of microbial fertilizer.
