摘要
为确定纤维素酶活力最高时的培养条件,研究了草菇菌株V23液体基本培养基的最佳碳源浓度与培养时间。结果表明:在微晶纤维素为碳源的情况下,微晶纤维素质量体积浓度为1%时草菇V23的纤维素酶活力最高;在1%微晶纤维素浓度下,培养第5天时草菇V23的纤维素酶活力达到最高值。在此基础上,对产量不同的草菇菌株V23和A8进行相同条件培养,分别测定其菌丝生长速度与纤维素酶活力。结果表明:在棉籽壳培养基中V23的菌丝生长速度显著快于A8;V23的纤维素酶活力也显著高于A8。
In order to explore cultural conditions to achieve the highest activity of cellulase,this paper studied the optimal concentration of microcrystalline cellulose and incubation time in liquid bas-al medium of Volvariella volvacea .The results showed that when the concentration of microcrystalline cellulose was 1 %,cellulase activity of V23 was the highest.Under this concentration of microcrystal-line cellulose,V23 had the highest activity of cellulase on the 5th day of incubation time.Strains V23 and A8 with different yields were cultured under the above condition and the growth speed and en-zyme activities were measured,respectively.V23 mycelium grew significantly faster than A8 on cotton hulls medium;the enzyme assay results showed that cellulase activity in V23 was significantly higher than that in A8.
