摘要
目的:探讨白子菜提取物对糖尿病小鼠血脂代谢的影响及对肝脏的保护作用。方法:高糖、高脂饲料饲养昆明小鼠,在第4周以链脲佐菌素(STZ)诱导Ⅱ型糖尿病小鼠模型,将造模成功的小鼠随机分为模型组、二甲双胍组、白子菜总提取物高、低剂量组和醋酸乙酯部位高、低剂量组,继续高糖高脂饲料喂养并连续灌胃给药4周后,检测各组小鼠体质量、空腹血糖值、三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白(LDL-c)、肝组织超氧化物歧化酶(SOD)和丙二醛(MDA)水平,并进行口服糖耐量试验(OGTT);HE染色后光镜观察肝脏组织的病理变化。结果:与模型组相比,各给药组均可明显促进糖尿病小鼠体质量的恢复,降低肝脏指数和肾脏指数,抑制血糖值升高,降低血糖曲线下面积,降低TG、TC、LDL-c水平,显著提高肝脏SOD活性,降低肝脏MDA水平,改善肝细胞形态,而醋酸乙酯部位的疗效优于总提取物。结论:白子菜提取物具有改善糖尿病小鼠血脂及一定的肝脏保护作用,其肝脏保护作用机制可能是通过增强肝组织抗氧化酶的活性、减少脂质过氧化物实现的。
OBJECTIVE To investigate the effects of Gynura divaricata(L.)DC.extracts(GDE)on serum lipid levels in diabetic mice and its therapeutic action on hepatic damage.METHODS Kunming mice were fed with high sugar and high lipid food for 4 weeks,and then intraperitoneally injected streptozotocin(STZ)to induce typeⅡ diabetes.The diabetic mice were randomly divided into model control group,metformin group,low and high dose of Gynura divaricata(L.)DC.ethanol extract groups,low and high dose of Gynura divaricata(L.)DC.ethyl acetate extract groups.After treated for 4 weeks,body weight,levels of blood glucose,triglyceride(TG),cholesterin(TC),low density lipoprotein cholesterol(LDL-c)were determined and oral glucose tolerance test was also performed.The activity of superoxide dismutases(SOD)and malondialdehyde(MDA)in liver tissue were detected.Morphological change was observed by HE staining.RESULTS Compared with model group,levels of blood glucose,the area under curve of the blood glucose,liver index,kidney index,TG,TC,LDL-c and MDA obviously decreased in each group treated with Gynura divaricata(L.)DC.extracts,while SOD activity and the body weight significantly increased,HE staining showed that Gynura divaricata(L.)DC.extracts had moderating effects on liver injury,and the ethyl acetate extract groups had better therapeutic effect.CONCLUSION These results suggested that Gynura divaricata(L.)DC.extracts can regulate lipid metabolism and provide protective effect on liver function of diabetic mice,and the mechanism of the liver protection may be related to the improved antioxidant activity and the decreased lipid peroxidation in the liver tissue.
