摘要
以三文鱼骨为原料,利用两步酶解法制得海洋骨胶原低聚肽粉,对其基础理化成分、氨基酸组成和分子量分布进行了分析,从DPPH自由基、羟基自由基、超氧阴离子自由基清除能力以及还原能力4个方面评价其抗氧化活性。理化特性分析表明,蛋白质含量为89.42%,氨基酸组成丰富,富含抗氧化性氨基酸;分子量小于1 000 Da的组分高达85.95%,132 Da^576 Da范围内组分占60.46%。抗氧化评价结果显示,海洋骨胶原低聚肽具有较强的抗氧化活性,并且呈量效关系,对DPPH自由基、羟基自由基和超氧阴离子自由基的半抑制质量浓度(IC50)分别为6.9 mg/mL,4.2 mg/mL和9.8 mg/mL,质量浓度为6.0 mg/mL时的还原能力与0.01 mg/mL的抗坏血酸相当。
Marine bone collagen oligopeptides(MBCP) was prepared from salmon bone by the method of two-step enzymolysis. Protein content, amino acid composition and molecular weight distribution of MBCP were determined. Antioxidant activities of MBCP were evaluated from four respects by determination of the scavenging activities against DPPH radical, hydroxyl radical and superoxide radical as well as reducing power. Physicochemical characterization analysis indicated that the content of total protein was 89.42% and the amino acids were abundant. MBCP were rich in antioxidative amino acids. The major molecular weight of MBCP was less than 1 000 Da(85.95%), and the peptides with a molecular weight range of 132~576 Da were 60.46%. The results of antioxidant evaluation showed MBCP exhibited antioxidant activities and the antioxidant activities showed a dose-response relationship with the concentration. The median inhibitory concentrations(IC50) of DPPH radical, hydroxyl radical and superoxide radical scavenging activities were 6.9 mg/mL, 4.2 mg/mL and 9.8mg/mL, respectively. And the reducing power of MBCP at 6.0 mg/mL was identical with that of ascorbic acid at 0.01 mg/mL.
