贯叶金丝桃药材HPLC指纹图谱研究

摘要 目的建立贯叶金丝桃HPLC指纹图谱分析方法。方法采用高效液相色谱法,色谱柱为DiamonsilC18(250 mm×4.6 mm,5μm)柱,流动相为乙腈(含10%甲醇和0.2%甲酸)、水(含20%甲醇和0.2%甲酸)梯度洗脱,检测波长为270 nm,流速为1.0 mL/min,柱温为室温。结果建立了贯叶金丝桃的指纹图谱,确定了13个共有峰,14批样品中有13批样品的指纹图谱与对照指纹图谱相似度在0.9以上,可用于贯叶金丝桃药材的定性鉴别。结论此方法简单、准确、重现性好,为贯叶金丝桃的质量标准研究奠定了基础。 Objective To establish a method of fingerprint analysis on Hypericum performatum L. by HPLC. Methods The HPLC method was used, and chromatographic conditions were as follow:Diamonsil C18 column (250 mm ×4. 6 mm,5 μm) ;acetonitrile( contain 10% methyl alcohol and 0.2% methanoic acid) -water( contain20% methyl alcohol and 0.2% methanoic acid) system as mobile phase at flow rate 1.0 mL/min; detective wavelength at 270 nm;column temperature was room temperature. Results The fingerprint of Hypericum performatum L. was established. 13 common peaks were found in the HPLC fingerprint of Hypericum performatum L.. Among 14 batches of samples,good similarities with correlation coefficients over 0.9 were found in the fingerprints between the 13 batches of samples from different sources and standard fingerprint, which could be utilized for the identification of Hypericum pelformatum L.. Conclusion The method is accuate, simple and good reproducibility and laid the foundation for the quality standard of Hypericum performatum L..
