PB实验设计联用CCD-RSM优化虎眼万年青多糖铁复合物的制备工艺

Optimization on preparation of ornithogalum caudatum ait polysaccharide iron complex based on Plackett-Burman design combined with CCD response surface methodology

摘要 为了探究虎眼万年青多糖铁(Ⅲ)复合物(OPIC)的最佳合成方法,我们采用Plackett-Burman设计对虎眼万年青多糖铁复合物制备工艺的主要影响因素进行考察,联用星点设计(central composite design,CCD)效应面法对显著性因素的水平进行优化,以溶剂加入量、三氯化铁加入量、多糖和柠檬酸钠加入量的比例、温度以及pH值为自变量,以虎眼万年青多糖铁中铁含量为因变量,通过对自变量与因变量的完全二次响应曲面的回归拟合,用效应面法预测最佳工艺.结果表明:温度、pH值和质量比对虎眼万年青多糖铁的合成有显著性意义;二项式方程拟合度高,相关系数r=0.987 9.虎眼万年青多糖铁的最佳合成工艺:溶剂的加入量为70mL,三氯化铁的加入量为6mL,温度80℃,质量比为3.5,pH值为8.最终利用PlackettBurman实验设计连用CCD确定了虎眼万年青多糖铁的最佳合成工艺,该方法简便,预测性好,重现性高. Object to choose the optimal synthesis method of Ornithogalum caudatum Ait polysaccharide iron complexes (OPIC). The influential factors of preparation technology of OPTC were investigated by Plackett-Burman design and optimized significant factors and level with the central composite de- sign. The independent variables were dosage of solvent, dosage of ferric chloride, the ratio of polysac- charide and dosage of sodium citrate (mass ratio), temperature and pH. The dependent variable was the OPIC iron content. We find that temperature, pH, and mass ratio had significant influence on the craft. Binomial,equation fitted high, the correlation coefficient reached 0. 987 9. The optimum condi- tions for the preparation of the complex were follows: the dosage of solvent was 70 mL, the dosage of ferric chloride was 6 mL, the mass ratio was 3.5, the temperature was 80 ℃, and the pH was 8. U- sing the Plackett-Burman design combined with CCD response surface methodology establish the opti- mal process of Ornithogalum caudatum Ait polysaccharide iron and the method is simple, good pre- dictability, and high reproducibility.
