摘要
从采集自温泉及高温堆肥的样品中分离纯化得到11株嗜热菌,通过分离菌株16SrDNA序列的分析对其进行了初步鉴定,其中地衣芽孢杆菌(Bacillus licheniformis)6株,凝结芽孢杆菌(Bacil luscoagulans)2株,嗜热脱氮芽孢杆菌(Geobacillus thermodenitrificans)1株,史氏芽孢杆菌(Bacil lussmithii)1株,嗜热脂肪芽孢杆菌(Geobacillus stearothermophilus)1株。并且针对蛋白酶、酯酶、淀粉酶、乳糖酶(β-半乳糖苷酶)、β-葡聚糖酶、木聚糖酶、羧甲基纤维素酶、α-半乳糖苷酶这8种饲料酶,采用唯一碳源或显色反应的方式对这些嗜热菌进行了产酶特性初步研究,检测结果表明这些嗜热菌均具有较强大的酶系,地衣芽孢杆菌和史氏芽孢杆菌产酶较多,分别可产其中的7种和6种酶。为嗜热酶的开发利用提供一定的理论基础。
Eleven thermophilic microorganisms were isolated from hot springs and compost samples, and identified as six Bacillus licheniformis strains, two Bacillus coagulans strains, Geobacillus thermodenitrificans, Bacillus smithii and Geobacillus stearothermophilus by 16 S rDNA analysis. And eight kinds of feed enzymes were detected by the method of single carbon source or chromogenic reaction for the isolated thermophiles including protease, esterase, amylase, β-galactosidase, β-glucanase, xylanase, CMCase and α-galactosidase. The results showed all the strains had rich enzyme systems, and Bacillus licheniformis and Bacillus smithii synthetized more kinds of these enzymes than others, 7 and 6 kinds respectively. Theoretical basis was provided for the exploitation of thermophilic enzyme.
