摘要
目的探讨感染对糖尿病(DM)大鼠模型病理状态的影响。方法雄性SD大鼠100只,随机分为两组,正常对照组10只,实验组90只,实验组采用链脲佐菌素(streptozotocin,STZ)65mg/kg一次腹腔注射方式建立DM大鼠模型,注射STZ后选取达到成模标准的大鼠作为DM模型组。12周时留取各组大鼠的肾脏、肺及血液标本,处死前一天,分别监测各组大鼠的进食量、饮水量、尿量、体重、空腹血糖(fasting blood glucose,FBG)、尿常规、24小时尿蛋白定量、肾功能、肾脏肥大指数(kidney hypertrophy index,KHI)、肾脏损伤分数。对收取的组织标本进行苏木素伊红染色(hematoxylin and eosin stain,HE)。根据肺脏及肾脏病理和尿常规等指标判断是否存在感染,对剔除感染前后的各项指标进行统计分析。结果 DM的成模率为58.89%,死亡率为43.33%。正常对照组大鼠无感染的发生,DM组大鼠感染的发生率为35.7%,其中感染大鼠的死亡率为80%。与正常对照组相比,DM组的进食量、尿量、饮水量、FPG水平、肌酐及尿素氮水平、尿常规中的红细胞及白细胞计数、24小时尿蛋白定量、KHI、系膜增生程度以及小管间质损害程度评分均显著增加,体重水平较低,具有统计学差异(P<0.05);与剔除感染前组相比,剔除感染后DM组大鼠的进食量较高,KHI、肌酐及尿素氮水平、尿常规中的红细胞以及白细胞计数、系膜增生程度评分以及小管间质损害程度评分均较低,具有统计学差异(P<0.05);与剔除感染前组相比,剔除感染后DM组大鼠的FPG水平及24小时尿蛋白定量水平较低,但不具有统计学意义(P>0.05)。结论感染可影响DM大鼠模型肾功能及肾脏病理改变的程度,为正确解读DM大鼠肾脏病变,在研究中需要剔除感染大鼠。
Objective To study the effects of infection on pathology in diabetic rats.Methods 100 male SD rats were randomly divided into control group(n=10)and experimental group(n=90).Experimental group were treated with intraperitoneal injection of STZ 65mg/kg once to establish the model of DM.After the injection of STZ,the rats reached to the modeling standards were selected as diabetes mellitus group.Kidney,lung and blood samples were harvested at the week of 12.One day before the rats were killed,the level of food intake,water intake,urine volume,body weight,fasting blood glucose,urine routine,24-hour urinary protein excretion,renal function,kidney hypertrophy index and kidney damages scores of rats of all groups were monitored.The pathological changes of tissue collected were observed by hematoxylin-eosin staining.While assess the index of kidney damage in rats of all groups.Infection was determined according to the pathology of lung and kidney and urine routine,all indicators were made for statistical analysis before and after the removal of infection.Results The rate of developed model of DM was 58.89% and the mortality rate was 43.33%.Infection was not occurred among the rats of control group.Infection rate of DM rats was 35.7%,but the mortality rate of DM infected model rats was 80%.Compared with control group,the food intake,water intake,urine output,FBG,creatinine,blood urea nitrogen,white blood cell and red blood cell count of urine routine,24-hour urinary protein excretion,KHI,degree of mesangial proliferation and tubulointerstitial damage scores of rats of diabetes mellitus were significantly higher and the body weight was significantly lower(P0.05).Food intake of diabetic rats after the removal of infection was significantly higher,the KHI,creatinine,blood urea nitrogen,white blood cell and red blood cell count of urine routine,degree of mesangial proliferation and tubulointerstitial damage scores of diabetic rats after the removal of infection was significantly lower compared with that of before the removal of infection(P0.05).FPG of diabetic rats after the removal of infection was slightly lower compared with that of before the removal of infection,but it does not have statistically difference(P0.05).The weight of diabetic rats were significantly lower compared with the control group(P0.05).The level of KHI、creatinine、blood urea nitrogen 、white blood cell and red blood cell of urine routine of rats of diabetes mellitus after the removal of infection were significantly lower compared with that of before the removal of infection(P0.05).FPG and 24-hour urinary protein excretion were lower,but it does not have statistical significance(P0.05).Conclusion Renal function and the degree of renal pathological changes can be affected by the infection,the rats of infection need to be removed in the study to correctly interpret the kidney lesions of DM rats.
