摘要
目的探讨双缩脲试剂在检测蛋白质应用中,不同检测条件对测试结果的影响。方法采用双缩脲法,选取三种不同仪器、试剂、校准物进行排列组合,组成27套检测系统,每个检测组合系统对5分样本血清进行检测,得到5个测定值,以全部血清总蛋白的测定值剔除离群值后得出的均值,以此计算出各个检测组合系统运用双缩脲试剂检测蛋白质的偏倚。结果不同检测仪器运用双缩脲试剂检测蛋白质的偏倚,方差齐性(P=0.467),差异不具有统计学意义(F=1.688,P=0.421);不同检测试剂运用双缩脲试剂检测蛋白质的偏倚,方差齐性(P=0.574),比较差异具有统计学意义(F=5.784,P=0.011);不同校准物运用双缩脲试剂检测蛋白质的偏倚,方差齐性(P=0.467),比较差异具有统计学意义(F=5.289,P=0.000)。结论双缩脲试剂在检测蛋白质应用中,检测试剂和校准物会对测试结果产生影响,在进行试验比对的时候,有必要对检测试剂和校准物造成的检测偏差进行考虑。
Objective To investigate the biuret reagent to detect proteins in the application, the impact of different test conditions for test results. Conclusion Biuret reagent in the detection of protein applications, impact detection reagents and calibrators will test result, during the test than when it is necessary to detect deviation detection reagents and calibrators due to be considered.
