nm23-H1转染对肺癌细胞钙调素和基质金属蛋白酶表达的影响

摘要 目的:探讨nm23-H1转染对肺癌细胞钙调素和基质金属蛋白酶表达的影响。方法:利用半定量RT-PCR技术检测转染前后E-cadherin、MMP-9基因的mRNA表达;利用流式细胞仪技术检测转染前后Ecadherin、MMP-9基因的蛋白表达。结果:转染后细胞株E-cadherin mRNA与蛋白表达上调,转染后细胞株MMP-9 mRNA与蛋白表达下调。结论:nm23-H1基因对肺癌细胞株L9981 E-cadherin mRNA与蛋白表达具有正调控作用,对MMP-9 mRNA与蛋白表达具有负调控作用。 Objective: To study influences of the transfection of nm23- H1 gene on the expression of E-cadherin and matrix metalloproterinases in lung cancer cell. Methods: Semi- RT- PCR was used to detect the mRNA expression of E- cadherin and MMP- 9 in transfected and non- transfected cell lines,Flow cytometry was used to detect the protein expression of E- cadherin and MMP- 9 in transfected and non- transfected cell lines. Results: The expression of mRNA and protein of E- cadherin of transfected cell line was stronger than that of non- transfected cell line. The expression of mRNA and protein of MMP- 9 of transfected cell line was weaker than that of non- transfected cell line. Conclusion: nm23- H1 can up- regulate expression of E- cadherin,down- regulate expression of MMP- 9 and thus inhibit the metastasis of lung cancer cell.
