摘要
目的:观察云药七龙天对缺氧大鼠的肺小动脉重构及肺组织中血管内皮细胞生长因子(VEGF)蛋白表达的影响。方法:SD大鼠40只随机分5组,正常对照组、模型对照组、七龙天低、中、高剂量组,以常压低氧法制模。第1天始灌胃,连续3周。行肺组织病理切片,观测肺小动脉重构情况:血管壁厚度、管壁占血管半径百分比(WT%)、血管壁面积、血管壁面积占血管面积百分比(WA%);采用蛋白免疫印迹法检测VEGF蛋白表达。结果:与正常对照组比较,各造模组肺小动脉重构指标均显著升高(P<0.05),VEGF蛋白表达显著升高(P<0.05);与模型对照组比较,七龙天各组上述指标水平下降,以七龙天高、中剂量组为显著(P<0.05)。结论:七龙天可显著改善缺氧性肺小动脉重构,其机制与降低VEGF蛋白表达,从而抑制缺氧诱导血管新生形成有关。
Objective: To observe the effects of Yunnan minority medicine Qilongtian on pulmonary arterioles remodeling and the protein expressions of vascular endothelial growth factor (VEGF) in the lung tissue of anoxic rats. Methods: 40 SD rats were randomly divided into 5 groups: the normal control group (C), the model control group (M), the low dose of Qilongtian group (QL), the medium dose of Qilongtian group (QM) and the high dose of Qilongtian group (QH). The atmospheric pressure and hypoxia environment was used to establish the anoxic rat model At the same time, corresponding medicines were gavaged for 3 weeks since the first day of modeling. The pulmonary arterioles remodeling, including the vessel wall, the percentage of vessel wall accounting for vascular radius (WT%), the acreage of vessel wall and its percentage accounting for vessel (WA%) were observed by pathological sections of lung tissues. The protein expressions of VEGF were detected by Western blot. Results: The pulmonary arterioles remodeling indexes and the protein expressions of VEGF in the model group were higher than those in the normal group (P〈0.05). The pulmonary arterioles remodeling indexes and the protein expressions of VEGF in each different dose of Qilongtian group were lower than those in the model group, especially in the QM and QH (P〈0.05). Conclusion: Qilongtian could significantly improve the pulmonary arterioles remodeling, the mechanism of which might be related to reduce the protein expressions of VEGF could inhibit the pulmonary angiogenesis induced by hypoxia.
