大豆异黄酮对异丙肾上腺素诱导大鼠心肌纤维化的保护作用 被引量:4

摘要 目的观察大豆异黄酮(Soybean isoflavone,SI)对异丙肾上腺素(Isoproterenol,ISO)诱导的心肌纤维化(Myocardial fibrosis,MF)的保护作用.方法异丙肾上腺素皮下注射诱导心肌纤维化模型,经SI低、中、高(30,60,120 mg/kg)3个剂量组和阳性对照卡托普利组(50 mg/kg)治疗后,观察其对心肌细胞直径(MD)、心室重量指数(HW/BW)、左心室重量指数(LVI)、羟脯氨酸(Hyp)含量及心肌组织中钙调神经磷酸酶(Calcineurin,Ca N)活性的影响.结果 SI不同剂量组均能够降低HW/BW和LVI(P<0.05或P<0.01),减少羟脯氨酸含量(P<0.05或P<0.01)),减小心肌细胞直径(P<0.05或P<0.01),降低心肌组织中Ca N的活性(P<0.01),减轻ISO诱导的心肌纤维化.结论 SI对心肌纤维化的保护作用可能与降低心肌组织中钙调神经磷酸酶活性有关. Objective To study the protective effects of soybean isoflavone ( SI ) on myocardial fibrosis induced by isoproterenol(ISO) in rats. Method Myocardial fibrosis model was induced by subcutaneous injections of ISO three groups of SI(30,60,120mg/kg),the change of cardio-myocyte diameter was measured by HE staining, cardiac mass index, left ventricular mass index ( LVI ) of rats were calculated after the end of experiment, hydroxyproline(Hyp) content,MD and calcineurin(CaN) activity in left myocardial tissue were quantified with spectrophotometry. Results Compared with ISO group, the heart weight index, LVI, Hyp content and CaN ( P〈0 . 01 ) activity in myocardial tissue were decreased obviously ( P〈0 . 05 or P〈0 . 01 ) . Conclusion The protective effects of soybean isoflavone on myocardial fibrosis is possibly related to decrease CaN activity in myocardial tissue.
