清热消积方对荷瘤鼠肿瘤组织HIF-1α蛋白表达的影响 被引量:4

摘要 目的观察清热消积方对肿瘤组织缺氧诱导转录因子-1α(HIF-1α)蛋白表达的影响,并探讨该方抗肿瘤血管生成的作用机制。方法将60只接种Lewis肺癌细胞株的C57BL/6J近交系小鼠随机分成空白对照组(NS组)、阳性对照组(CTX组)、清热消积方高、中、低剂量组(生药量1.12 g/m L、0.56 g/m L、0.28 g/m L),每组12只。空白对照组和清热消积方高、中、低3个剂量组按0.2 m L/10 g分别喂饲生理盐水和中药水煎液,阳性对照组腹腔注射环磷酰胺。用免疫组织化学法和免疫印迹法检测荷瘤小鼠肿瘤组织HIF-1α蛋白表达。结果清热消积方高、中、低剂量组、CTX组肿瘤组织HIF-1α蛋白的平均光密度(MOD)值均低于NS组(P<0.05),清热消积方高、低剂量组、CTX组肿瘤组织HIF-1α蛋白的阳性细胞所占值(A%)均低于NS组(P<0.05),清热消积方各剂量组间MOD值与A%均无差异(P>0.05)。结论清热消积方能下调荷瘤鼠肿瘤组织HIF-1α蛋白的表达,可能是其抗肿瘤血管生成的作用机制之一。 Objective To investigate the effect of Qing Re Xiao Ji on the expression of hypoxia inducing factor 1 alpha(HIF-1α)protein of Tumor Tissue,and to explore the mechanism of anti-tumor angiogenesis. Methods 60 C57BL/6J inbred mice with Lewis lung carcinoma xenograft model were randomly divided into five groups:blank control group(NS group),positive control group(CTX group)and Qing Re Xiao Ji group divided into high,medium and low dose group(the amount of crude drug were 1.12g/m L,0.56g/m L, 0.28g/m L), 12 rats in each group. The blank control group and Qing Re Xiao Ji high, medium, low dose group were respectively feed with physiological saline,traditional Chinese medicine decoction in 0.2m L/10 g,the positive control group was treated with intraperitoneal injection of cyclophosphamide. Using immunohistochemical method and Western blot to detect the expression of HIF-1α protein of Tumor Tissue. Results The MOD of HIF-1α protein in Qing Re Xiao Ji high,medium,low dose group,CTX group were lower than NS group(P〈0.05),the A% of HIF-1α protein in Qing Re Xiao Ji high,low dose group,CTX group were lower than NS group(P〈0.05),and Qing Re Xiao Ji in each dose group showed no difference between MOD and A%(P〉0.05). Conclusion Qing Re Xiao Ji can cut the expression of HIF-1α protein,which may be one of anti-tumor angiogenesis effects.
