右美托咪定预处理对大鼠肝门阻断所致肠道损伤的保护作用 被引量:1

Protective Effect of Dexmedetomidine Preconditioning on Intestinal Inj ury Induced by Portal Triad Clamping

摘要 目的探讨α2-肾上腺素能受体激动剂右美托咪定预处理对大鼠肝门阻断所致肠道损伤的保护作用。方法24只雄性 SD大鼠随机分为3组(n=8):假手术组(Sham组)、肝门阻断组(PTC组)、右美托咪定预处理组(Dex组)。应用Pringle法行肝门阻断,持续30 min,阻断解除后4 h 处死大鼠,取血清测肿瘤坏死因子-α(TNF-α)水平,取回肠组织测定超氧化物歧化酶(SOD)、丙二醛(MDA)水平,行病理镜检并作 Chiu评分。结果阻断解除后4 h,PTC组及Dex组血清TNF-α水平、回肠组织MDA含量及病理学Chiu评分均显著高于 Sham组,回肠组织 SOD活性显著低于 Sham 组;Dex 组 TNF-α、MDA 及 Chiu 评分结果显著低于PTC组,而 SOD结果显著高于PTC组(P< 0.05)。结论右美托咪定可通过减轻组织炎症及氧化应激反应,保护肠黏膜,减轻肝门阻断后肠道的损伤。 Obj ective]To explore the protective effect ofα2-adrenergic receptor agonist dexmedetomidine preconditioning on intestinal inj ury induced by portal triad clamping in rats.[Methods]Twenty-four SD male rats were randomly divided into sham operated group(Sham group,n=8),portal triad clamping group(PTC group,n=8)and dexmedetomidine preconditioning group(Dex group,n=8).The hepatic portal triad was clamped by Pringle maneuver and maintained for 30min.After 4h of clamping release,rats were killed.Serum level of tumor necrosis factor-alpha(TNF-α)was detected.Ileal tissue was collected to detect superoxide dis-mutase(SOD)and malondialdehyde(MDA).Pathological examination and Chiu scoring were performed.[Re-sults]After 4h of clamping release,the levels of serum TNF-α,MDA in ileal tissue and Chiu scores in PTC group and Dex group were markedly higher than those in Sham group,but the activity of SOD in ileal tissue was markedly lower than that in Sham group.The levels of TNF-α,MDA and Chiu scores in Dex group were markedly lower than those in PTC group,while the activity of SOD was markedly higher than that in PTC group(P〈 0.05).[Conclusion]Dexmedetomidine can protect intestinal mucosa and alleviate intestinal injury after portal triad clamping by reducing tissue inflammation and oxidative stress response.
