摘要
目的:观察骨痹消对佐剂性关节炎兔软骨中肿瘤坏死因子(tumor necrosis factor alpha,TNF-α)、白细胞介素1(interleukin-1,IL-1)水平的影响,探寻骨痹消治疗骨关节炎的作用机制。方法:取新西兰兔40只,雌雄各半,体质量450 g左右,其中10只为空白对照组,其余30只采用佛氏佐剂诱导兔关节炎模型。模型制作成功后,随机分为模型组、西药组、中药组,连续给药9d。中药组和西药组分别灌胃给予中药骨痹消水煎剂0.01 m L·g-1、罗非西布0.01 m L·g-1(0.2 mg),对照组和模型组灌胃给自来水0.01 m L·g-1,每天灌胃1次,常规饲养,给药9 d后取材,采用ELISA法测定兔关节软骨中TNF-α、IL-1含量。结果:中药组干预后兔关节软骨中TNF-α质量浓度为(19.52±1.84)μg·L-1、IL-1质量浓度为(54.95±7.24)ng·L-1,与正常组相比较,其余各组兔软骨中TNF-α、IL-1水平均显著升高(P<0.05);模型组兔软骨中TNF-α、IL-1水平显著高于中药组和西药组(P<0.05);但中药组兔软骨中TNF-α、IL-1水平显著低于西药组(P<0.05)。结论:骨痹消可抑制骨关节炎兔软骨细胞产生IL-1和TNF-α,这可能是骨痹消治疗佐剂性关节炎作用机制之一。
Objective: To observe Gubixiao on adjuvant arthritis rabbits cartilage tumor necrosis factor( tumor necrosis factor alpha,TNF-α),interleukin^-1( IL-1) level influence,and to explore Gubixiao mechanism for the treatment of osteoarthritis. Methods: 40 New Zealand white rabbits,male and female,body weight of about 450 g,of which only 10 into blank control group,the remaining 30 only use of Freund's adjuvant induced arthritis model in rabbits. The success of the model,were randomly divided into model group,western medicine group,Chinese medicine group,continuous administration of 9 d. Traditional Chinese medicine group and western medicine group respectively by intragastric administration of traditional Chinese medicine Gubixiao water decoction of 0. 01 mL ·g^-1,rofecoxib 0. 01 mL ·g^-1( 0. 2 mg),the model group and the control group by intragastric administration of water 0. 01 mL ·g^-1,gastric perfusion every day 1 times,conventional breeding,after administration of 9 days material,determination of TNF-α,IL-1 content in rabbits joint soft bone by ELISA method. Results: Traditional Chinese medicine group of dry TNF-α prognosis of articular cartilage is( 19. 52 ± 1. 84) μg·L^-1,IL-1( 54. 95 ± 7. 24) ng·L-1,compared with the normal group,the TNF-α level of IL-1 alpha and other groups in rabbit cartilage were significantly increased( P〈0. 05); TNF-α,IL-1 level of model group rabbit cartilage were significantly higher than those of the traditional Chinese medicine group and western medicine group( P〈0. 05); however,TNF-α,IL-1 level of Chinese herbal medicine group was significantly lower than that of rabbit cartilage in western medicine group( P〈0. 05). Conclusion: Gubixiao can inhibit rabbit osteoarthritis cartilage cells to produce IL-1 and TNF-α,which may be one of Gubixiao adjuvant arthritis treatment mechanism.
