摘要
以发根农杆菌Accc10060菌株侵染地黄外植体,研究不同菌液浓度、不同侵染时间和不同共培养时间对地黄毛状根诱导率的影响。结果表明,发根农杆菌Accc10060菌株可成功诱导出地黄毛状根,PCR检测发现发根农杆菌Ri质粒rol C基因已整合到地黄基因组中并得到表达;单因素试验结果显示,在地黄毛状根的诱导过程中,诱导的最佳菌液浓度为OD600=0.6,最佳侵染时间为10 min,最佳共培养时间为4 d。
Agrobacterium rhizogenes Accc10060 was used to infect the explants of Rehmannia glutinosa,then the influences of different bacterial concentrations,infection times and different co- culture times on hairy root induction rate were researched. The results showed that the hairy roots of R. glutinosa could be induced by A. rhizogenes Accc10060,and PCR detection results indicated that the rol C gene in Ri plasmid of A. rhizogenes had integrated into the genome of R. glutinosa and been expressed. The results of single factor experiment showed that the best induction conditions of hairy roots were OD600 as 0. 6,infecting for 10 minutes and co- culturing for 4 days.
