摘要
分别以乙醇和水为提取剂,提取桉树叶中的有效成分,观察其对7种耐甲氧西林金黄色葡萄球菌(MRSA)的抑菌作用,并将未加桉树提取物和加了桉树提取物的苯唑西林的抑菌作用进行对比。结果表明,桉树的乙醇提取物和水提取物对7种MRSA的最小抑菌浓度(MIC)分别为1-5g/L和2-10g/L。添加0.2g/L的桉树乙醇提取物之后,苯唑西林对7种MRSA的MIC范围由62.5-250mg/L减小至7.8-31.3mg/L,添加1g/L的桉树水提取物之后,苯唑西林对7种MRSA的MIC,除了试验菌株MRSA No.16之外,减小至7.8 mg/L,而MRSA No.16的最小抑菌浓度从250mg/L减小至62.5mg/L。试验结果证明了桉树提取物与苯唑西林对7种MRSA具有良好的协同抑菌效果,采用乳胶凝集法发现,桉树乙醇提取物抑制青霉素结合蛋白2(PBP2’)的合成,从而降低MRSA对苯唑西林的耐药性。
Ethanol and water are used respectively to extract the active ingredients of eucalyptus leaves.Then it was researched on the inhibitory effect of the active ingredients on MRSA,and compared the inhibitory effect of the oxacillin with and without eucalyptus extracts addition.The results shows that,the ethanol extract of eucalyptus presents a minimum inhibitory concentration(MIC)from 1g/L to 5g/L for MRSA,and the water extract is from 2-10g/L.With 0.2g/L of ethanol extract of eucalyptus added,the oxacillin can reduce the MIC of the MRSA from 62.5-250mg/L to 7.8-31.3mg/L,and the MIC for all kinds of MRSA reduce to 7.8mg/L except MRSA NO.16 which is reduced from 250mg/L to 62.5mg/L after adding in0.1% water extract.The fractional inhibitory concentration(FIC)index method is used to present that the eucalyptus extract and oxacillin have good synergic inhibitory effect against MRSA,and latex agglutination is used to find that,ethanol extract of eucalyptus inhibit the produce of PBP2'of MRSA and reduce the drug resistance of MRSA on oxacillin.
