槲皮素对重症急性胰腺炎相关性肺损伤肺泡中性粒细胞TLR4/NF-κB通路的影响 被引量:5

摘要 目的探讨槲皮素(Quercetin)对大鼠重症急性胰腺炎相关性肺损伤(SAP-LI)肺泡中性粒细胞(PMN)TLR4/NF-κB信号通路的影响。方法 48只成年SD大鼠随机分为4组,每组12只,假手术(Sham)组、重症急性胰腺炎(SAP)组、槲皮素低剂量组(50 mg/kg)及槲皮素高剂量组(100 mg/kg)。逆行性胰胆管注射5%牛磺酸钠建立SAP大鼠模型。槲皮素组在胰腺炎模型基础上予槲皮素干预,Sham组、SAP组给予等量生理盐水。计算肺湿/干重比(W/D)和肺通透指数(LPI),经支气管肺泡灌洗分离收集PMN,ELISA检测PMN培养上清液中TNF-α含量,RT-PCR检测PMN TLR4 m RNA表达,Western blotting检测PMN TLR4和NF-κB p65的表达。结果与Sham组比较,其余三组W/D、LPI及TNF-α含量明显升高,TLR4 m RNA及其蛋白和NF-κB p65蛋白表达上调;与SAP组比较,槲皮素处理组W/D、LPI及TNF-α含量降低,TLR4 m RNA及其蛋白和NF-κB p65蛋白表达下调。结论槲皮素减轻SAP引起的肺损伤可能与其抑制PMN TLR4/NF-κB信号通路活化有关。 Objective To explore the effects of Quercetin on Toll-like receptor 4/NF-κB signal pathway in alveoar PMN from rats with severe acute pancreatitis-associated lung injury(SAP-LI). Methods Forty-eight SD rats were randomly divided into four groups: Sham group, SAP group, Que low doses group(50 mg/kg) and Que high doses group(100 mg/kg). SAP model was induced via biliary pancreatic duct retrograde administration of 5%sodium taurocholate. Que group was given Quercetin on the basic of SAP model, while Sham group and SAP group were administrated with the same volume of saline. Calculate the lung wet/dry weight ratio(W/D) and lung permeability index(LPI). PMN were harvested by bronchoalveolar lavage. The content of TNF-α in the supernatant fluid of cultured PMN was detected by ELISA, expression of TLR4 m RNA in PMN was measured by RTPCR, Western blot analysis was used to test the expression of TLR4 and NF-κB p65 in PMN. Results Compared with Sham group, W/D, LPI and TNF-α content were markedly increased, expression of TLR4 m RNA as well as protein and NF-κB p65 in PMN were up-regulated in three other groups. Compared with SAP group, W/D, LPI and TNF-α content were decreased, expression of TLR4 m RNA as well as its protein and NF-κB p65 protein were down-regulated in Que treated groups. Conclusion Quercetin alleviate lung injury caused by SAP which was probably related to its inhibition of Toll-like receptor 4/NF-κB signal pathway in alveolar PMN.
