摘要
目的探讨大黄素对子宫内膜异位症(内异症)异位内膜细胞的干预效果。方法体外分离培养内异症异位内膜上皮及间质细胞,分别加入不同浓度(20、40、80、160μmol/L)大黄素进行干预。Annexin V FITC/PI流式细胞术检测细胞凋亡,结晶紫染色法检测细胞黏附,Transwell小室检测细胞侵袭。结果大黄素可诱导内异症异位内膜上皮及间质细胞凋亡,并抑制其侵袭(P<0.05),但未见明显剂量依赖性;大黄素可剂量依赖性抑制内异症异位内膜上皮及间质细胞黏附。结论大黄素可能单独或作为手术后辅助药物对内异症有一定的治疗作用,有可能成为治疗内异症的新选择之一。
Objective To evaluate the effects of emodin on ectopic endometrial cells of endometriosis. Methods Endometrial epithelial cells(EECs) and endometrial stromal cells(ESCs) in the endometriomas of patients with endometriosis were separated and cultured. The cultured cells were treated with different doses(20 μmol /L,40 μmol /L,80μmol/L and 160 μmol/L) of emodin. The apoptosis of cells was detected by flow cytometry with annexinV FITC/PI double staining. The adhesion of cells was detected by crystal violet staining. The invasive ability of cells was detected with transwell chambers. Results Apoptosis of EECs and ESCs was induced by emodin,which also significantly suppressed the adhesive and invasive capacities of EECs and ESCs( P〈0. 05). Conclusion Emodin may become a novel option for treatment of endometriosis.
