摘要 目的:探讨micro RNA-125b(mi R-125b)在口腔鳞状细胞癌(OSCC)患者血浆中的表达及临床意义。方法:用茎环法反转录(RT)-实时荧光PCR法检测85例OSCC患者和46例健康人血浆中mi R-125b的表达水平,评估其作为OSCC诊断标志的检测效能。采用SPSS17.0软件包中的单因素方差分析和ROC曲线分析对结果进行统计学处理。结果:反转录PCR法可特异性扩增血浆中的mi R-125b,与正常对照组相比,OSCC患者组血浆mi R-125b表达水平显著上调(P<0.05);ROC曲线分析显示,血浆mi R-125b水平可有效区分OSCC患者和正常人群,其曲线下面积(AUC)达到0.966;当确定最佳截断值为3.46(fold值)时,血浆mi R-125b对OSCC的诊断敏感度和特异度分别达到89.4%和93.5%。结论:鉴于其良好的诊断效能,血浆mi R-125b有望成为具有潜在应用价值的OSCC生物学标志物。 PURPOSE: To explore the expression and clinical significance of plasma micro RNA-125b(mi R-125b) in patients with oral squamous cell carcinoma(OSCC). METHODS: By Stem-loop RT real-time PCR based on the SYBR Green, we investigated the expression of plasma mi R-125 b in 85 OSCC patients and 46 healthy controls, then evaluated its diagnostic performance as OSCC biomarker. Data analysis was performed with SPSS 17.0 software package for one-way ANOVA and receiver operating characteristic(ROC) curve. RESULTS: Stem-loop RT-PCR could specifically amplify mi R-125 b in plasma. The expression of plasma mi R-125 b was significantly increased in patients with OSCC compared with healthy controls(P0.05). ROC results showed that mi R-125 b could differentiate OSCC from healthy controls(AUC=0.966). As fold change 3.46 was chosen for optimal cutoff value, the diagnostic sensitivity and specificity was 89.4% and93.5%, respectively. CONCLUSIONS: Due to the satisfactory diagnostic performance, plasma mi R-125 b can be used as a promising biomarker in OSCC.
