长期起居饮食不节致亚健康状态大鼠模型的研究 被引量:3

Experimental study on the model of sub-healthy rats caused by long-term irregular life schedule and disorder diet

摘要 目的:模拟现代人"起居无常、饮食失节"致生物钟紊乱现象,对不节律生活致亚健康大鼠模型进行探索。方法:打破大鼠正常生活节律,给予大鼠长期24h/24h间歇睡眠剥夺联合饮食失节法造成亚健康状态。采用单数日24h起居自由+高糖高脂饲料(过食肥甘)、偶数日24h睡眠剥夺+白菜(单一素食)方法造模。比较正常组与模型组大鼠行为学指标、血液学指标、心电学的差异,并观察模型可逆性。结果:与正常组比较,1行为学:模型组自造模第2周出现耳赤现象、抓力显著降低,自第4周痛阈显著降低,自第5周面温显著升高;停止造模1周后面温恢复正常水平、痛阈、抓力无明显改善。2血常规:自造模第5周模型组白细胞(WBC)水平显著降低,血小板分布宽度(PDW)、大血小板比率(P-LCR)显著升高,停止造模1周后血小板分布宽度(PDW)及大血小板比率(P-LCR)恢复正常,白细胞水平(WBC)未见明显恢复。3血生化:自造模第5周血糖(GLU)、总胆固醇(TC)显著增高,甘油三酯(TG)显著降低;停止造模1周后血糖水平(GLU)及甘油三酯水平(TG)恢复正常,总胆固醇(TC)未见明显恢复。4心电:模型组P波高度明显上升,R波高度、QRS波群宽度显著上升,平均心电轴右偏(QRSa);5环核苷酸系统及细胞因子:模型组大鼠血浆c AMP水平及c AMP/c GMP比值显著升高,IL-2水平显著降低,IL-6水平显著升高。结论:采用长期间歇睡眠剥夺联合饮食失节复合因素造模可令大鼠出现耳赤、易惊、乏力等体虚症状,并使大鼠血糖血脂代谢紊乱、血液学指标及心电波形异常、细胞因子和环核苷酸系统紊乱,与人体亚健康表现较接近,可为补益中药干预亚健康等研究提供新的思路或方法。 Objective: To simulate modern people "irregular life schedule and disordered diet" resulting in disrupted biological clock, explore and evaluate the model of the sub-healthy rats caused by irregular life style. Methods: Use long-term 24h/24h interval sleep deprivation com- bined with diet disorder method to break the normal life rhythm in rats. The detailed method is as follows: On odd-numbered days, model rats live a 24h normal life and on a high-sugar plus high fat diet ( overeating sugar and fat), on even-numbered days use 24h sleep deprivation and feed model rats Chinese cabbage (only vegetarian food). Then compare the differences between the normal group and the model group in the areas of hematology, behavioristies, eleetroeardiology, and observe the reversibility of the model. Results: Compared with normal rats, ①In behavioristics area: After modeling 2 weeks, model rats appeared flushed ears phenomenon, rats'holding power signifieandy reduced (P 〈 0.01 ) ; After modeling 4 weeks, model rats'pain threshold significantly reduced ( P 〈 0.01 ) ; After modeling 5 weeks, model rats'facial temperature significantly rised ( P 〈0.01 ) ; Stop modeling one week, model rats'facial temperature went back to normal, while holding power and pain threshold didnt obviously recover. ②In blood routine test: After modeling 5 weeks, the level of white blood cell(WBC) count sig- nificantly reduced ( P 〈 0.01 ), platelet distribution width(PDW) significantly increased ( P 〈 0.05 ), platelet-large cell rate (P-LCR) signif- icantly increased (P 〈 0.01 ) ; Stop modeling one week, model rats'platelet distribution width (PDW) and platelet-large cell rate (P-LCR) went back to normal, while the level of white blood cell (WBC) count didnt obviously recover. ③ In blood biochemistry test: After modeling 5 weeks, blood glucose(GLU) and total cholesterol(TC) significantly increased ( P 〈 0.01 ), while triglyceride(TG) significantly decreased (P 〈0.01 ) ; Stop modeling one week, model rats'blood glucose (GLU) and triglyeeride(TG) went back to normal, while total cholesterol (TC) didnt obviously recover. ④In ECG test: Model rats'amplitude of P wave significantly increased ( P 〈0.01 ), amplitude of R wave sig- nificantly increased ( P 〈 0.05 ) , width of QRS wave group significantly increased ( P 〈 0.05 ), and appeared right axis shift of the mean e- lectrical axis(QRSa) (P 〈0.01 ). ⑤ In cytokines area: Model rats'plasma cAMP and eAMP/cGMP ratio significantly increased (P 〈 0. 01 ) , IL-2 level significantly reduced ( P 〈 0.01 ) , IL-6 level significantly increased ( P 〈 0.01 ). Conclusion: Irregular life style of "irregu- lar life schedule and disordered diet" will lead to sub-health status in rats, such as deficiency syndrome ( flushed ears, susceptible to scare and feebleness, ere) , blood glucose and blood fat metabolism disorder, abnormal hematology index and ECG waveform. After modeling 4 weeks, it appeared significant differences between model group and normal group, and the differences continued to 9th week. The model had reversibility to some degree. The study show that using long-term interval sleep deprivation combined with diet disorder method can built the sub-healthy model in rats.
