摘要
[目的]揭示不同产地金樱子30个居群间的遗传多样性和亲缘关系。[方法]采用RAPD分子标记技术,从35条RAPD随机引物中,筛选出8条有效引物进行扩增,用NTSYSpc,ver.2.02软件进行UPGMA聚类分析。[结果]共扩增出86条带,其中多态性条带84条,多态性百分率(PPB)97.67%。30个居群的相似性系数在0.11-0.58,聚成A、B、C三大类群。[结论]金樱子具有丰富的遗传多态性,产自贵州地区的金樱子有较近的亲缘关系。
[Objective] This essay is devoted to find out the genetic diversity and phylogenetic relationship of thirty provenances of Rosa laevigata Michx from different areas. [Method] The software of‘NTSYSpc,ver. 2. 02'was adopted to conduct UPGMA analysis on eight samples which were chosen from 35 random primers by RAPD. [Result]According to the research,86 loci appeared,including 84 polymorphic ones,which told a polymorphic percentage of 97. 67%. This study also found the similarity coefficients from 0. 11 to 0. 58 of Rosa laevigata Michx.Based on this data,those 30 populations were divided into 3 groups as‘A',‘B'and‘C'. [Conclusion] Rosa laevigata Michx is a kind of species with multiple genetic polymorphism. Those Rosa laevigata Michx from Guizhong Province have close genetic relationships.
