摘要
目的探讨从枸杞中分离得到的甜菜碱对异丙肾上腺素(ISO)诱导的大鼠急性心肌缺血损伤的保护作用机制。方法大鼠分别灌胃给予不同剂量甜菜碱(100、200和400 mg/kg)共40 d,于第39天和第40天皮下注射ISO85 mg/(kg·d)诱发大鼠急性心肌缺血。取大鼠心肌组织进行病理学观察,ELISA法测定血清TGF-β1与ET-1的水平,并采用Western-blot法测定相关蛋白的表达。结果与模型组比较,甜菜碱(100、200和400 mg/kg)明显改善心肌组织坏死,炎细胞浸润及间质水肿。甜菜碱(100、200、400 mg/kg)可明显降低血清TGF-β1及ET-1的水平(P<0.01),抑制心肌组织Rho A、ROCK1、ROCK2蛋白表达的升高(P<0.01)。结论甜菜碱可通过调控Rho A/ROCK信号通路保护ISO诱导的大鼠急性心肌缺血损伤。
AIM To investigate the cardioprotective effects of betaine extracted from Lycium barbarum L.on regulation of Rho A / ROCK pathway in isoproterenol-induced acute myocardial ischemia.METHODS Betaine( 100,200 and 400 mg /kg) was administered orally once a day for forty days,and acute myocardial injury was induced by subcutaneous injection of isoproterenol( 85 mg / kg) in rats on the 39 th and 40 th day.Myocardial injury was examined by histopathological evaluation.Serum levels of transforming growth factor( TGF)-β1 and endothelin( ET)-1 were analyzed by ELISA.In addition,the protein expression of Rho A、ROCK1and ROCK2 were measured by Western-blot.RESULTS Oral administration of betaine( 100,200 and 400 mg / kg) attenuated myocardial necrosis,interstitial edema,and inflammatory cell infiltration.Moreover,the treatment with betaine decreased isoproterenol-induced elevated serum levels of TGF-β1 and ET-1( P〈0.01) as well as increased protein expression of Rho A,ROCK1 and ROCK2( P〈0.01).CONCLUSION The present study has demonstrated that betaine protects against isoproterenol-induced myocardial ischemia via down-regulating Rho A / ROCK pathway.
